Abstract

Artan dünya nüfusu nedeniyle insan beslenmesinde temel besin maddelerinin başında gelen buğdaya olan gereksinim hızlı bir şekilde artmaktadır. Ancak buğday üretimi verim kayıpları nedeniyle yeterli miktarda olmamaktadır. Buğday üretiminde ürün veriminin düşük olmasının önemli nedenlerinden biri abiyotik stres faktörlerinin başında gelen kuraklıktır. Bu nedenle buğday çeşitleri ve hatlarında kuraklığa tolerans bakımından genetik karakterizasyon çalışmalarının yapılması, kuraklığa dayanıklılık bakımından genetik sınıflandırma ile öne çıkan genotiplerin tespit edilmesi ve yapılacak ıslah çalışmalarında kullanım potansiyeli olan genotiplerin belirlenmesi önem teşkil etmektedir. Bu tez kapsamında kuraklıkla bağlantılı farklı gen bölgeleriyle ilişkili moleküler markörler kullanılarak buğday genotiplerinin kuraklık toleransı açısından genetik karakterizasyonları yapılmıştır. Bu amaçla kullanılan kontrol grupları arasında polimorfik olan ve net ayırım sağlayan 6 adet SSR ve 9 adet ISSR olmak üzere 15 adet moleküler markör kullanılmıştır. Çalışma sonucunda kuraklığa toleranslı genotiplerin sınıflandırılması gerçekleştirilmiş ve öne çıkan genotipler belirlenmiştir. İncelenen 27 adet ekmeklik buğday genotipinin 9 adet ISSR markörü ile genetik karakterizasyonu sonucunda % 53.4 polimorfizm oranı tespit edilmiş, ortalama PIC değeri 0.60 olarak hesaplanmıştır. Altı SSR markörüyle yapılan moleküler taramalar sonucunda ise ortalama PIC değeri 0.77 olarak hesaplanmış olup, heterozigotluk oranı 0.79 ve ortalama allel sayısı ise 6.7 olarak belirlenmiştir. Çalışma sonucunda kuraklığa toleranslı ve hassas genotiplerin sınıflandırılması gerçekleştirilmiş ve öne çıkan genotiplerin daha sonra yapılacak olan ıslah çalışmaları için kullanım potansiyelleri ortaya konulmuştur.

Due to the increasing world population, the need for wheat, which is one of the main nutrients in human nutrition, is rapidly increasing. However, wheat production is not sufficient due to yield losses. Drought is one of the major causes of abiotic stress factors in wheat production. Therefore, genetic characterization studies in terms of drought tolerance in wheat varieties and lines, determination of tolerant genotypes with genetic classification in terms of drought resistance, and thus identification of genotypes that have potential to be used in breeding studies are important.In this thesis, genetic characterizations of wheat genotypes in terms of drought tolerance have been made by using molecular markers related to different gene regions related to drought. For this purpose, 15 molecular markers were used, 6 SSRs and 9 ISSRs, which are the most polymorphic and provide clear discrimination among the control groups. As a result of the study, the classification of drought tolerant genotypes was realized and the prominent genotypes for future breeding studies were determined. As a result of genetic characterization of 27 bread wheat genotypes with 9 ISSR markers, 53.4% polymorphism rate was determined and the mean PIC was calculated as 0.60. In the genetic characterization performed with six SSR markers, the mean PIC value was 0.77, the heterozygosity was 0.79 and the mean allele number was 6.7. As a result of this study, the classification of drought tolerant and sensitive genotypes was realized and the potentials of the prominent genotypes for future breeding studies were revealed.
