Abstract

Tilki kuyruğu (Ceratophyllum demersum L.) Ceratophyllaceae familyasına ait bir su bitkisidir. Su ortamındaki ağır metallerin uzaklaştırılmasında (fitoremediasyon) ve kirliliğin izlenmesinde (biyomonitör) yaygın olarak kullanılmaktadır. Bazı çiftlik hayvanları ve kümes hayvanları için iyi bir besin kaynağıdır. Ayrıca, akvaryumlarda ticareti yapılan popüler bitkiler arasında yer almaktadır. Bu çalışma, su ekosisteminde ve ticari açıdan önemli olan C. demersum'un in vitro koşullarda çoğaltımı için hazırlanmıştır. Bitkinin yüzey sterilizasyonu için çeşitli konsantrasyon ve sürelerde hidrojen peroksit (H2O2) ve çamaşır suyu (NaOCI) kullanılmıştır. En fazla steril ve sağlam eksplantlar, %5'lik H2O2 ile 7 dk muamele ile elde edilmiştir. Sürgün rejenerasyonu amacıyla sürgün ucu, 1. ve 2. koltukaltı meristem eksplantları farklı konsantrasyonlarda veya kombinasyonlarda sitokininleri (BAP, TDZ ve Kinetin), oksinleri (IBA ve NAA) veya gibberellik asiti (GA3) içeren agarla katılaştırılmış ya da sıvı MS besin ortamlarında kültüre alınmıştır. Agarla katılaştırılan kültür ortamlarında eksplant başına sürgün sayısı en fazla (61,92 adet) 0,10 mg/l TDZ, 0,10 mg/l IBA ve 0,10 mg/l GA3 kombinasyonlarını içeren MS ortamında sürgün ucu meristem eksplantından elde edilmiştir. Sıvı kültür ortamlarında ise en fazla (204,33 adet) sürgün 0,40 mg/l BAP içeren MS ortamında 2. koltukaltı meristem eksplantından elde edilmiştir. Her iki kültür ortamlarından elde edilen rejenere bitkilerin su ortamına başarıyla adaptasyonu sağlanmıştır.Anahtar Kelimeler: Adaptasyon, Ceratophyllum demersum, Çoğaltım, Doku Kültürü, In Vitro, Sterilizasyon, Sürgün Rejenerasyonu

Coontail (Ceratophyllum demersum L.) is an aquatic plant belongs to Ceratophyllaceae family. The plant has been widely used to remove heavy metals (photoremediation) and pollution monitoring (biomonitor) in the aquatic environment. It is also a good food source for livestock and poultry. Furthermore, it is one of the popular plants in aquatic industry. The present study was designed for in vitro propagation of C. demersum, which is commercially valuable and important for water ecosystem. Hydrojen peroxide (H2O2) and commercial bleaching agent (NaOCI) were used at various concentrations and times for surface sterilization. Maximum sterile and live explants were obtained from 5% H2O2 with aplication time of 7 minutes. Shoot meristem, 1st and 2nd node explants were cultured on agar solidified or liquid MS medium supplemented with different concentrations or combinations of cytokinins (BAP, TDZ and Kin), auxins (IBA and NAA) or gibberellic acid (GA3) for shoot regeneration. Maximum number of shoots per explant (61,92) was obtained from shoot meristem on agar solidified MS medium containing 0,10 mg/l TDZ, 0,10 mg/l IBA and 0,10 mg/l GA3. Whereas, maximum number of 204,33 shoots was achieved on liquid MS medium supplemented with 0,40 mg/l BAP from 2nd node explant. The regenerated plants obtained from both cultures media were succesfully acclimatised in aquatic environments.Keywords: Acclimatization, Ceratophyllum demersum, In Vitro, Propagation, Shoot Regeneration, Sterilization, Tissue Cultere
