Patatesin (Solanum tuberosum) abiyotik stres tepki mekanizmasındaki MAPK sinyal iletim ağı

Abstract

Abiyotik stres, tüm bitkilerde olduğu gibi patates içinde önemli verim kayıplarına sebep olmaktadır. Evrimsel olarak korunan ve abiotik stres tepki mekanizmasında da çok önemli bir yere sahip olan MAPK sinyal ağı birçok bitkide çalışılmasına rağmen patates ile ilgili sınırlı çalışma mevcuttur. Tez kapsamında, 1) abiyotik stres ile ilişkili StMAPK'ler belirlenmiş, 2) miRNA'lar ile etkileşimleri ve 3) transkripsyon faktörleri ile etkileşimleri in-siliko olarak incelenmiştir. Elde edilen sonuçlara göre; 4 adet yüksek sıcaklık, 3 adet tuz ve 4 adet kuraklık stresi tepki mekanizmasında görevli muhtemel MAPK sinyal ağı tespit edilmiştir. StMAPKKK16-1/2/3/4/5/6, StRaf1, StMAPKKK4-1/2, StMAPKKK21-1/2, StMKK3-2, StMKK5, StMKK9, StMPK3-2, StMPK4-2, StMPK8-3, StMPK9-2, StMPK16-2, StMPK19-4 ve StbHLH (PGSC0003DMP400016023)'in patates abiyotik stres tepki mekanizmasında aktif görev aldığı saptanmıştır. Elde edilen sonuçlar ile abiyotik stres tepkisinde MAPK sinyal ağı in siliko olarak aydınlatılmıştır. Bu çalışma doğrultusunda elde edilen çıktıların ileride deneysel olarak kanıtlanmasıyla, abiyotik stres tepki mekanizması için oldukça önemli olan MAPK sinyal iletim ağı kesin olarak aydınlatılmış olacaktır.
