Efecto inductivo in vitro de citoquininas sobre el brotamiento de segmentos nodales de palo de rosa (Aniba rosiodora Ducke)

Abstract

The present research work was carried out at the Biotechnology Laboratory of the National Institute for Agrarian Innovation (INIA), located in the district of Juan Guerra, in the San Martin region. The objective was to evaluate the inductive effect of three sources and concentrations of cytokinins on the sprouting of Brazilian rosewood (Aniba rosiodora) nodal segments under in vitro conditions. Vegetative segments and M&S culture medium supplemented with: T1 (KIN - 0.45 μM); T2 (KIN - 2.32 μM); T3 (KIN - 23.23 μM); T4 (BAP- 0.45 μM); T5 (BAP - 2.32 μM); T6 (BAP - 23.23 μM); T7 (2Ip - 0.45 μM); T8 (2Ip - 2.32 μM); T9 (2Ip - 23.23 μM) were used. They were distributed in totally randomized designs with a 3A (Sources) x 3B (Concentrations) factorial arrangement and a control treatment. Ten treatments and eight replicates were obtained. In order to reduce the incidence of pathogenic microorganisms in the culture medium, preventive applications were made in the field with a fungal solution at a rate of 1 gL-1, 30 days before each extraction, together with the addition of Vitrofural at the culture medium level; the segments were sterilized with a solution of 2% NaClO + tween 80 for 20 minutes. The data obtained were submitted to analysis of variance and the Tukey multiple range test (p < 0.01). It was determined that by applying preventive applications in the field 30 days before in vitro installation, a survival rate of more than 88 % was reached. Likewise, T5 (BAP- 2.32 μM) and T6 (BAP-23.23 μM) reached a higher number of shoots and high averages with respect to shoot length, obtaining a shoot length of (8.41 mm and 6.13 mm) respectively. Finally, it was identified that T9 (2Ip - 23.23 μM) and T8 (2Ip - 2.32) optained better average in callus formation with 1.2 callus. It is concluded, that the BAP cytokinin source induces greater shoot activation in Brazilian rosewood timber species at the in vitro level.

El presente trabajo de investigación se ejecuto en el Laboratorio de Biotecnología del Instituto Nacional de Innovación Agraria (INIA), ubicado en el distrito de Juan Guerra, región San Martín. El objetivo consistió en evaluar el efecto inductivo de tres fuentes y concentraciones de citoquininas sobre el brotamiento de segmentos nodales de palo de rosa (Aniba rosiodora) in vitro. Se empleó segmentos vegetativos y un medio de cultivo M&S suplementado con: T1 (KIN - 0,45 μM); T2 (KIN - 2,32 μM); T3 (KIN - 23,23 μM); T4 (BAP- 0,45 μM); T5 (BAP – 2,32 μM); T6 (BAP – 23,23 μM); T7 (2Ip - 0,45 μM); T8 (2Ip - 2,32 μM); T9 (2Ip - 23,23 μM). Se distribuyeron en diseños totalmente aleatorizados con arreglo factorial 3A (Fuentes) x 3B (Concentraciones) y un tratamiento control. Se obtuvieron 10 tratamientos y 8 repeticiones. Para reducir la incidencia de microorganismos patógenos en el medio de cultivo, se realizaron aplicaciones preventivas en campo con una solución fúngica a razón de 1 gL-1, 30 días antes de cada extracción, sumado a la adición de vitrofural a nivel del medio de cultivo; los segmentos se esterilizaron con una solución de NaClO al 2 % + tween 80 durante 20 minutos. Los datos obtenidos fueron sometidos al análisis de varianza y la prueba de rango múltiple Tukey (p <0,01). Se determinó que realizando aplicaciones preventivas en campo 30 días antes de la instalación in vitro logramos tener una supervivencia mayor al 88 %. Asimismo, el T5 (BAP- 2,32 μM) y T6 (BAP-23,23 μM) lograron un mayor número de brotes y promediós altos con respecto a la longitud del brote, logrando una logitud de (8,41 mm y 6,13 mm) respectivamente. Finalmente se identificó que los T9 (2Ip - 23,23 μM) y T8 (2Ip - 2,32) optuvieron mejor promedio en la formación de callo con 1,2 callos. Se concluye, que la fuente de citoquinina BAP induce una mayor activación de brotes en la especie forestal palo de rosa a nivel in vitro.
