
V ÖZET ATIK SULARDA FOSFOR GİDERİMİ YAPAN BAKTERİLERİN İNCELENMESİ PAŞAYEVA, Pikehanım Yüksek Lisans Tezi, Biyoloji Bölümü Tez Yöneticisi: Prof. Dr. Rengin ELTEM Temmuz 2003, 63 sayfa Polifosfat bakterileri İzmir İzsu Biyolojik Atıksu Arıtım Tesisi'nin aerobik ve anaerobik fosfat giderim havuzlarından üç farklı tarihte alınan atık su örneklerinden izole edilmişlerdir. Fosfat gideren çeşitli bakterilerin pH 6,5' da fosfat giderim verimliliklerinin karşılaştırılması, fosfat gideriminde etkili olan Gr(-) bakterilerin, özellikle, de Acinetobacter spp.'nin tanımlanması amaçlanmıştır. İzolasyonda 'Acetat Defined Medium' (ADM), 'Baumann Zenginleştirme' ortamı ve 'Glyserol Casiton Yeast Extrakt' (GCYP) ortamları kullanılmıştır. Bu ortamlar kullanılarakelde edilen 91 saf bakteri izolatına Gram boyama, oksidaz, katalaz, testleri yapılmıştır. İzolatların laboratuvar koşullarında fosfat giderim kapasiteleri ölçülmüş, içlerinde en iyi fosfat giderimi yapan 11 Gr (-) bakteri seçilerek API 20 NE identifikasyon kitleri ile tür seviyesinde tanımlanmıştır. Tanısı yapılan türlerden 8 'inin Acinetobacter baumanii/calcoaceticus, 3 'ünün de Pseudomonas aeruginosa olduğu belirlenmiştir. VII ABSTRACT A SURVEY ON THE BACTERIA EFFECTIVE IN THE WASTEWATER PHOSPHORUS REMOVAL PAŞAYEVA, Pikehanim Msc in Biology Department Supervisor: Prof.Dr. Rengin ELTEM July 2003, 63 pages Samples were extracted from the aerobic and anaerobic phosphate removal tanks of the mil-scale municipal biological wastewater treatment plant îzsu in Izmir City at three different dates and polyphosphate bacteria were isolated from this samples. The comparison of phosphate removal efficiencies of various phosphate accumulating bacteria at pH 6,5, the identification of more effective phosphate removers, i.e. Gr(-) bacteria and especially Acinetobacter spp. were aimed. ADM (Acetate Defined Medium), GCYP (Gliserol Casiton Yeast Extract), Baumann Enrichment Medium were used for isolations and 91 isolates isolated. Gram staining, oxidase and catalase tests were applied to all isolates. Phosphate removal ability of this bacteria were measured in laboratory conditions and 11 most efficient Gr (-) bacteria were selected. Identification studies were done by using identification kit; API 20 NE. Based on the results obtained, eight isolates as Acinetobacter baumanii/calcoaceticus and three isolates as Pseudomonas aeruginosa identificated.
