Abstract

ÖZET FİTAZ ENZİMİ ÜRETİMİ VE OPTİMİZASYONU ÖZATAY, Şehnaz Yüksek Lisans Tezi: Gıda Mühendisliği Bölümü Tez Yöneticisi: Prof. Dr. Süha SUKAN Ocak, 2001, 99 sayfa Bu tezde, ilk olarak, karbon kaynağı olarak seçilmiş buğday kepekli üretim ortamında 9 çeşit mikroorganizmadan fitaz enzimi üretebilenler belirlenmiş ve bu mikroorganizmalardan maksimum aktivitede fitaz enzimi üretebilen Aspergillus ficuum, Aspergillus terreus (M) ve Aspergillus carbonarius seçilmiştir. Bu seçilen mikroorganizmalar ile üretilen fitaz enzimi aktivitesi üzerine besin elementi ve dışarıdan fosfat ilavesinin etkileri incelenmiştir. Başlangıç ortam pH'sı, zaman ve azot kaynağının aktivite üzerine etkilerini görebilmek amacıyla; 4 farklı başlangıç pH'sında, 3 farklı zaman aralığında, 5 değişik azot kaynağı kullanılarak yapılan üretimler sonucunda en yüksek aktiviteyi veren mikroorganizma seçilmiştir. En yüksek aktivitede fitaz enzimini kısa sürede elde edebilmek amacıyla misel inokülasyonunun ve inokülasyon yüzdesinin etkileri de incelenmiştir. vn ABSTRACT PRODUCTION AND OPTIMIZATION OF PHYTASE ENZYME ÖZATAY, Şehnaz MSc in Food Engineering January 2001, 99 pages In this study, 9 microorganisms were grown on the medium containing wheat bran as carbon source and the phytase activities of these microorganisms were determined. The highest activities were optained with Aspergillus ficuum, Aspergillus terreus (M) and Aspergillus carbonarius. The effects of nutrients and phosphate addition on the phytase activity of these organisms were determined. To determine the effect of initial pH of the medium, production time interval and nitrogen source on the phytase activity; microorganisms were grown on medium with 4 different initial pH and 5 different nitrogen sources and 3 different time intervals. To produce phytase at higher activity in an optimum period of time, the effect of micelium concentration and inoculation percentage on enzyme activity were also examined.
