Abstract

IV ÖZET EGE VE DO?U KARADENİZ BÖLGESİ TOPRAKLARINDAN İZOLE EDİLEN AKTİNOMİSETLER'DEN ANTİBAKTERİYAL ANTİBİYOTİKLERİN ARANMASI VE ÜRETİMİ ÜZERİNE BİR ARAŞTIRMA DENİZCİ Aziz Akın Doktora tezi, Biyoloji Anabilim Dalı Tez Yöneticisi: Prof. Dr. Mehmet Öner Eylül 1996, 148 sayfa Ege ve Doğu Karadeniz topraklarından 356 aktinomiset kültürü izole edilmiştir. İzole edilen 356 aktinomiset kültürlerinden 128 kültürün antibiyotik aktivite gösterdikleri saptanmıştır. Şöyleki; bunlardan 114 izolat sadece Gr (+) bakterilere, 1 izolat sadece Gr(-) bakterilere, 13 izolat da hem Gr(+) bakterilere hem de Gr(-) bakteriler üzerine etkin antibiyotik ürettikleri görülmüştür. Aktivite ve spektrum durumları dikkate alınarak 356 Aktinomiset kültürü arasından seçilen 4 izolattan, izolat K26-10 Streptomyces exfoliatus ; izolat K7-12 Streptomyces lydicus ; izolat K26-1 Streptomyces albidoflavus ; izolat K29-5 Streptomyces nojiriensis olarak tanılanmışlardır. Seçilen 4 izolatın herbiri ile yapılan fermentasyon çalışmalarında, bütün izolatlar soya fasulyesi unu kullanılan ortamda (Besiyeri 28) en yüksek antibiyotik üretimini gerçekleştirmişlerdir. Üretilen antibiyotiklerin peptit yapıda olmadıkları, ikisinin kuvvetli polar diğer ikisinin apolar oldukları saptanmıştır. Üretilen antibiyotiklerden üçü preparatif ince tabaka kromatografisi (TLC) ile saflaştınlmışlardır. Anahtar kelimeler: Aktinomisetler, izolasyon, identifikasyon ve antibiotik üretimi.

V ABSTRACT A STUDY ON THE DETECTION AND PRODUCTION OF ANTIBACTERIAL ANTIBIOTICS FROM ACTINOMYCETES WHICH ISOLATED FROM THE SOILS OF AEGEAN AND EASTERN BLACK SEA REGIONS OF TURKEY DENİZCİ Aziz Akin PHD in Biology Supervisor: Prof. Dr. Mehmet Öner September 1996, 148 pages 356 Actinomycetes cultures were isolated from soils of Aegean and Eastern Blacksea regions. Antibiotic activity were shown by 128 of 356 isolated cultures. So that, 114 isolates produce antibiotic against only on Gram(+) bacteria, 1 isolates only against Gram(-) bacteria and 13 isolates against both Gram(-) and Gram(+) bacteria. In accordance with antibacterial activity and spectrum broadness four isolates were selected and identified among 356 Actinomycetes cultures as below: The isolate K26-10 as Streptomyces exfoliatus; the isolate K7-12 as Streptomyces lydicus; the isolate K26-1 as Streptomyces albidoflavus; the isolate K29-5 as Streptomyces nojiriensis. In the studies of fermentation studies by using 4 selected isolates, all isolates have showed highest antibiotic activity in the medium (Medium 28) containing soy bean meal. None of the 4 producing antibiotic were not shown peptid structure. On the other hand, two of them were strongly polar and the other two were apolar. Three of producing antibiotics were purified by using preparative thin layer chromotography(TLC). Keywords: Actinomycetes, isolation, identification and antibiotic production.
