Abstract

-21- SUMMARY In this study a prosedüre for the isolation and culture of protoplast of 6/7 and Granola genotypes grown in greenhouse ba sed on the metoda of Haber lach et al (1985) was used. Excised leaves of plants were preconditioned for 2 days in the dark at 4*G and 1 day at CON media. Eight and 16 hour enzyme digestion followed with purification by flotation in Babcock bottles. Washed protoplasts were counted on a hemocytometer and the pro toplast yields were determined. The following results were obtained : 1. There was a variation among the genotypes in terms of protoplast yield. 2. Protoplast yields changed depending on genotype, enzyme digestion time and plant age. 3. 6/7 genotype was found to be superior to the Granola genotype in terms of protoplast yield. 4. The highest protoplast yield were found to be at 6 week old plants. 5. 16 hour enzyme digestion time found to be suitable as compared to the 8 hour incubation time. Based on the results obtained in this experiment, the use of 5-6 week old 6/7 plant, an 16 hour enzyme digestion make this prosedüre both convenient and efficient.

.20- Ö2ET Bu çalışmada; 5,6,7 haftalık 6/7 ve Granola patates çeşit lerine ait 1 g.' lık yaprak örnekleri, Haberlach ve ark., (1985) 'nın metoduna bağlı kalarak muamele edilmiştir. Bu amaçla, yaprak örnekleri 2 gün karanlıkta buzdolabında ve sonra 1 gün CON ortamında tutulduktan sonra, Q ve 16 saat süre ile enzimde inkUbe edilmiştir. SUre bitiminde, protoplast sayımı hemosito- metre ile yapılarak protoplast verimleri tespit edilmiştir. Çalışma sonucunda elde edilen sonuçlar sırasıyla: 1. Kullanılan genotipler arasında protoplast verimi bakı mından farklılıklar vardır. 2. Protoplast verimi genotip, enzim muamele süresi ve yaprak yaşma bağlı olarak değişmektedir. 3. 6/7 klonu, Granola çeşidine göre daha yüksek protoplast verimine sahiptir. 4. En yüksek protoplast verimi 6. inci haftada elde edilmiştir. 5. 16 saatlik enzim inkübasyonu, 8 saatlik inkübasyona göre daha başarılı sonuçlar vermiştir.
