Abstract

Bu çalışmada, polietilen glikol (PEG) 6000 ile uyarılan kuraklık stresine maruz kalan Lotus corniculatus çeşidinde fotorespirasyonun rolü; büyüme parametreleri, bağıl su içeriği (RWC), prolin, malondialdehit (MDA) birikimi, membran sızıntısı, yaprak ozmotik potansiyeli, stoma iletkenliği (gs), fotorespirasyon oranı (PR), net fotosentetik verim (Fv/Fm), fotosentetik CO2 asimilasyon oranı (A), içsel CO2 konsantrasyonu (Ci), fotokimyasal olmayan enerji dağılımı (NPQ), elektron taşıma oranı (ETR) ve transpirasyon oranları (E) bakımından araştırılmıştır. Ayrıca kuraklığın fotorespirasyon metabolizmasına etkisi fotorespirasyonel döngüde yer alan fosfoglikolat fosfataz (PGP), katalaz (CAT), glutamat: glioksilat aminotransferaz (GGT) gibi enzimlerin biyokimyasal analizleriyle belirlenmiştir.İki farklı PEG konsantrasyonu uygulanan L. corniculatus, % 10 PEG'e göre % 20 PEG' ten daha fazla etkilenmiştir. Kuraklık stresine bağlı olarak L. corniculatus'ta büyüme parametreleri ve RWC' de azalma gözlenirken, prolin ve MDA birikiminde, membran sızıntısında, yaprak ozmotik potansiyelde artış tespit edilmiştir. Kuraklık stresi koşullarında L. cornciulatus' ta gs, Fv/Fm, A, ETR ve E' nın azaldığı, Ci, NPQ ve PR' nın arttığı belirlenmiştir. L. corniculatus bitkisinde artan NPQ ve PR değerleri, fotosentetik aparatların kuraklık stresine karşı fotorespirasyon ve mehler-peroksidaz yolakları tarafından korunduğunu göstermektedir. Kuraklık stresinin, PGP ve CAT enzim aktivitelerini azalttığı gözlenirken, GGT enzim aktivitesini önemli derecede arttırdığı saptanmıştır. Ayrıca kuraklık stresinde, GGT enzim aktivitesi ve fotorespirasyon oranı arasında bağlantının olduğu gözlenmiştir.Elde edilen bu bulgular, L. corniculatus çeşidinde kuraklık stresi boyunca fotorespirasyonun arttığını ve fotorespirasyonel döngünün kuraklık stresine verilen yanıtların düzenlenmesinde önemli rol oynayabileceğini göstermektedir.

İn this study, the role of photorespiration in Lotus corniculatus cultivar subject to dorught stress induced by polyethylene glycol (PEG) 6000 was studied in view of growth parameters, relative water content (RWC), proline, malondialdehyde contents (MDA), membrane ion leakage, leaf osmotic potential, stomata conductance (gs), photorespiration rates (PR), net photosynthetic efficiency (Fv/Fm), photosynthetic CO2 assimilation rates (A), intercellular CO2 concentrations (Ci), non-photochemical quenching (NPQ), electron transport rates (ETR), transpiration rates (E). Additionally, the effect of drought on the photorespiration metabolism was determined by biochemical analyze such as activities of some photorespiratory cycle enzymes such as phosphoglycolate phosphatase (PGP), catalase (CAT) and glutamate: glyoxylate aminotransferase (GGT). L. corniculatus of applied two difirent PEG concentrations was affected % 20 PEG more than % 10 PEG. Growth parameters and relative water content (RWC) decresaed but proline, MDA, membrane ion leakage, leaf osmotic potential increased in L.corniculatus under drought stress. Fv/Fm, gs, A, ETR, E decresaed and Ci, NPQ, PR increased in L. corniculatus under drought stress. İncrease of NPQ and PR indicate that photorespiration and Mehler-peroksidaz pathways protects photosynthetic apparatuses in L. corniculatus plant against drought stress. While PGP and CAT enzyme activties decreased under drought stress, GGT enzyme activity remarkably increased. Furthermore, correlation between GGT enzyme and PR was observed under drought stress. These findings demonstrate that photorespiration was enchanched during drought stress and that photorespiratory cycle might have a role in regulate responses to drought stress in L.corniculatus cultivar.
