Abstract

Bal arıları olarak da bilinen Apis mellifera türü, Dünya’da ekolojik açıdan oldukça önemli bir yere sahiptir. Dünya’da yaygın olarak bulunan bu sosyal böcekler, diğer canlı organizmalara ev sahipliği yapmaktadır. Bakteri, virüs, fungus ve akarlar gibi organizmalar, bal arılarından faydalanarak, arıların bağışıklık sistemini zayıflatarak arılarda bazı hastalıklara sebebiyet vermektedir. Arılarda görülen hastalıklar ve koloni kayıpları, arı verimini düşürmesiyle birlikte ekolojik hayatı tehlikeye sokmaktadır. Bu durumun ortadan kaldırılması için bazı mücadele yöntemleri kullanılmaktadır. Fiziksel, kimyasal ve organik maddeler gibi mücadele yöntemlerinin kullanılmasıyla birlikte bu parazitlerle mücadele sürecinde kullanılan ürünlerin arılarda kalıntı bırakması, uygulanan dozun ayarlanamamasından dolayı kovanlardaki kolonilerin zarar görmesi, ekonomik açıdan maliyetin düşürülmesi gibi birçok sebeple yeni mücadele yöntemlerine ihtiyaç duyulmaktadır. Bu tez çalışması kapsamında arıların gastrointestinal sisteminden izole edilen ve probiyotik olarak kullanılabilme potansiyeline sahip oldukları bilinen izolatlar tanılanarak, bioassay yöntemi ile Varroa destructor türü akarlara karşı biyolojik mücadele yöntemi ile mikrobiyal biyokontrol ajanlarının araştırılması amaçlanmaktadır. Yapılan çalışmalar sonucunda, 16SrDNA dizi analizleri yapılan ve tanılanan bu 3 izolatların Bacillus amyloliquefaciens (C11A ve C12B) ve Bacillus velezensis (C12A1) olduğu belirlenmiştir. Bioassay denemesi sonucunda potansiyel probiyotik özellik gösterebilme yeteneğine sahip 3 izolattan, (C12A1) B. velezensis türünün akarların hareketliliğini %50 oranında, diğer 2 izolatın ise (C11A, B. amyloliquefaciens) %10-%30, (C12B, B. amyloliquefaciens) %30-%40, elde edilen 3 izolattan oluşturulan konsorsiyumun ise akarların hareketliliğini %30 oranında inhibe ettiği görülmüştür.

Apis mellifera, also known as honey bees, has an ecologically important place in the world. These social insects, commonly found on Earth, are home to other living organisms. Organisms such as bacteria, viruses, fungi and mites cause some diseases in bees by taking advantage of honey bees and weakening the immune system of bees. Diseases and colony losses in bees endanger ecological life, as well as reducing bee productivity. Some methods of struggle are used to eliminate this situation. With the use of control methods such as physical, chemical and organic substances, new control methods are needed for many reasons such as the products used in the fight against these parasites leave residue on the bees, the damage to the colonies in the hives due to the inability to adjust the applied dose, and the reduction of economic costs. Within the content of this thesis, it is aimed to identify the isolates isolated from the gastrointestinal tract of bees and known to have the potential to be used as probiotics, and to investigate microbial biocontrol agents with the bioassay method and the biological control method against Varroa destructor type mites. As a result of the studies, it was determined that these 3 isolates, whose 16SrDNA sequence analyzes were performed and identified, were Bacillus amyloliquefaciens (C11A and C12B) and Bacillus velezensis (C12A1). As a result of the bioassay experiment, of the 3 isolates capable of showing potential probiotic properties, the motility of the (C12A1) B. velezensis species was 50%, the other 2 isolates (C11A, B. amyloliquefaciens) 10%-30%, (C12B, B. amyloliquefaciens) %30-%40. It was observed that the consortium formed from 3 isolates inhibited the motility of the mites by 30%.
