Abstract

Mikro-Ribonükleik Asit ler (miRNA), ~ 22 nükleotid uzunluğunda bulunan, bitkiler, hayvanlar ve virüslerde gen ekspresyon ağlarında anahtar düzenleyiciler olarak birçok biyolojik prosesi etkileyen protein kodlamayan RNA lardır. miRNA lar keşfedildikleri zamandan itibaren medikal, biyolojik ve ziraat bilimlerinde önemli gelişmelere sebep olmuşlardır. Et üretimi için önemli bir çiftlik hayvanı olan koyun aynı zamanda biyolojik ve karşılaştırmalı genomik çalışmaları için de ideal bir model organizmadır. Çalışma perspektifine göre yeni miRNA dizisi belirleme yöntemleri, deneysel metodlar veya bilgisayar temelli teknikler olarak değişebilmektedir.Bu tez çalışmasında, hem deneysel yöntemler hem de biyoinformatik yaklaşımlar uygulanarak koyun genomundan transkribe edilen miRNA dizilerinin belirlenmesi hedeflenmiştir. Bu amaçla koyun kanından elde edilen kısa-RNA?lara ait cDNA dizileri klonlanarak analiz edilmiş ve 1 adet olası aday miRNA dizisi bulunmuştur. Ayrıca, uygulanan homoloji temelli biyoinformatik yöntemde mevcut sığır miRNA?ları kullanılarak 63 homolog aday miRNA dizisi tahmin edilmiştir. Toplam olarak 64 adet olası miRNA saptanmıştır.

Micro-Ribonucleic Acids (miRNA) ~ 22 nt. long and non-protein coding RNAs, effect various biological processes as key regulators of gene expression networks in plants, animals and viruses. Since their discovery, miRNAs induce significant developments in medical, biological and agricultural sciences. Sheep an important agricultural livestock for meat production, is also an ideal model organism for biological and comparative genomics researches. Experimental methods and computational technics are the approaches can be changeable according to perspective of the study for identification of novel miRNA sequences.In the present study, identification of miRNA sequences transcribed from sheep genome by both experimental methods and bioinformatics approaches were aimed. With this purpose, cDNA sequences of the small-RNAs obtained from sheep blood were cloned and analyzed. One candidate miRNA sequence was identified. Also, by using known cattle miRNAs in homolgy-based bioinformatics methods, 63 homologous candidate miRNA sequences were predicted. Totally, 64 potential miRNAs were determined.
