Abstract

Bitki mitokondri organelinin en önemli görevlerinden biri bitkiye enerji kaynağı olarak ATP sağlamasıdır. ATP kısaca, organik asitlerin oksidasyonuyla oluşan elektronların, solunum elektron transferi zinciri (ETS) ile oksijene transfer olması sonucu meydana gelir. Ayrıca mitokondri önemli ikincil fonksiyonlara da sahiptir. Bunlar; nükleotidlerin sentezlenmesi, aminoasit ve lipid metabolizmaları, vitamin ve kofaktörlerin sentezlenmesi, ışık solunum yolağına katılması ve organik asit ara ürünlerinin daha geniş hücresel biyosentezi için taşınmasıdır.Bu çeşitli görevlerin üstesinden gelebilmesi için mitokondri 1000 ila 1500 farklı protein içermektedir ve bu proteinlerin bitkideki karmaşık fizyolojik ve genetik işlevlerinin aydınlatılmasında proteomik analizler son 10 yılda güçlü bir araç haline gelmiştir.Çalışmamızda, 100 mM konsantrasyonda NaCl uygulanmış Nicotiana tabacum bitkisinin mitokondrisi izole edilmiştir ve mitokondri protein profili 2D PAGE ile belirlenmiştir. Ayrıca, western blot analizi ile mitokondrinin izole edildiği teyit edilmiştir. Protein profilleri sonuçlarında, kontrol grubu ile karşılaştırılan uygulama grubunun protein profillerinde tuz stresine bağlı değişim tespit edilmiştir. Ek olarak; tuz stresinin 1., 3. ve 7. günündeki örneklemelerin analizleri, protein profilindeki değişiminin NaCl stresinin süresine bağlı olduğunu göstermiştir.

One of the most important tasks of mitochondria is to provide ATP to plants as an energy source. Respiratory oxidation of organic acids and the transfer of electrons to O2 via the respiratory electron transport chain are coupled to the synthesis of ATP. Mitochondria also performs many important secondary functions related to synthesis of nucleotides, metabolism of amino acids and lipids, synthesis of vitamins and cofactors, participation in the photorespiratory pathway, and export of organic acid intermediates for wider cellular biosynthesis.To undertake these diverse roles, mitochondria contain between 1000 and 1500 different proteins and proteome analyses became a powerful tool for the investigation of complex genetic and physiological processes for the last 10 years.In this study, we aimed to investigate the differences in mitochondrial proteome of salt stressed Nicotiana tabacum plants. We isolated mitochondria from 0 and 100 mM NaCl treated plants on days 1 and 7. After testing the intactness and purity of mitochondria by western blotting we performed 2D PAGE for mitochondria proteins. Protein profiles differed in salt stressed groups as compared to that of control groups. The quantities and intensities of the protein spots were changed in salt stresses groups during the experimental period.
