Abstract

Çalışmamızda ilk aşamada mikoparazitik aktiviteye sahip olduğu önceden saptanmış ve moleküler tanılaması yapılmış olan beş Trichoderma izolatı ile bir ticari Trichoderma harzianum suşunun farklı inkübasyon sıcaklıklarında koloni büyümesi ve spor oluşumları incelenmiştir. Ayrıca modifiye Czapek Ortamına (MCM) eklenen 16 farklı karbon ve 8 farklı azot kaynağının, izolatların koloni büyüme hızlarına ve spor oluşumlarına etkisi değerlendirilmiştir. Daha sonra temel karbon kaynağı olarak % 0,5 oranında kitin içeren modifiye ?Trichoderma Liquid Enzyme Production Medium? (TLE Ortamı)'da iki farklı inkübasyon sıcaklığında 4 ve 7 gün süreyle büyütülen izolatlarda kitinaz, ß-1,3-glukanaz ve proteaz aktiviteleri incelenmiştir. Tüm izolatlar çeşitli yönlerden (litik aktivite, büyüme sıcaklık aralığı, sporlanma dereceleri ve önceden saptanmış olan mikoparazitik aktivite) incelenerek biyokontrol ajanı olabilecek etkinlikteki suş olarak Trichoderma citrinoviride P25 belirlenmiş ve bu suş katı kültür fermentasyonu üretim optimizasyonu çalışmalarında kullanılmıştır.Çalışmamızda ikinci aşamada, substrat olarak buğday kepeğinde büyütülen T. citrinoviride P25'in öncelikle erlenlerde daha sonra da paslanmaz çelik tepsilerde üretim optimizasyonu çalışması yapılmıştır. Katı kültür ortamına azot kaynağı olarak malt çimi % 5,0 oranında eklendiğinde en yüksek mikropropagül sayısına ulaşılmıştır. Katı kültür ortamının optimum başlangıç pH değeri (4,5) ve bu ortama yapılacak optimum inokulum miktarı (1,0x108 spor/ml) belirlendikten sonra, yanıt yüzeyi yöntemi (RSM) kullanılarak optimum inkübasyon sıcaklığı (27,46 oC), başlangıç nem oranı (% 68,80) ve inkübasyon süresi (6,89 gün) de belirlenmiştir. Daha sonra ölçek büyütme amacıyla tepsilerde aynı koşullar altında farklı substrat miktarları (84, 105, 126, 147, 168, 189, 210 ve 262,5 g) denenmiş ve en yüksek toplam mikropropagül sayısının (2,96x1012 kob/262,5 g katı substrat) elde edildiği substrat miktarı (262,5 g) ile tepsilerde üretim gerçekleştirilmiştir.

Firstly in our study, five molecularly identified mycoparasitic Trichoderma isolates and one commercial Trichoderma harzianum strain were incubated at different temperatures and colony growth and spore production of these isolates were investigated. Also, the effect of 16 different carbon and 8 different nitrogen sources added to modified Czapek Medium (MCM) on colony growth and spore formation rate of isolates were evaluated. Afterwards, isolates cultured in Trichoderma Liquid Enzyme Production Medium (TLE) containing 0.5% chitin as a sole carbon source, were incubated at two different temperatures for 4 and 7 days and chitinase, ß-1,3-glucanase and protease activities of isolates were studied. All of the isolates were evaluated in respect to lytic activities, growth temperature ranges, sporulation degrees and predetermined mycoparasitic activities. In this way, the strain capable of a biocontrol agent was determined and Trichoderma citrinoviride P25 strain was chosen for optimization studies with solid-state fermentation.In the next stage of our study, optimization of production conditions for T. citrinoviride P25 strain cultured on wheat bran as a substrate was studied at erlenmayer flasks and stainless steel trays respectively. Highest micropropagule number was achieved in the medium containing 5% malt sprout as a nitrogen source. After the optimal initial pH of the solid medium (pH 4.5) and inoculum size (1.0x108spore/ml) were determined, optimal values of incubation temperature (27.46 oC), initial moisture rate (68.80 %) and incubation time (6.89 days) were determined by using response surface methodology (RSM). With the aim of scaling up, different substrate amounts (84, 105, 126, 147, 168, 189, 210 and 262.5 g) were tested in trays under same conditions and production was carried out with substrate amount (262.5 g) which provides highest total micropropagule number (2.96x1012 cfu/262.5 g solid substrate).
