Abstract

Fasulye (Phaseolus vulgaris L.) içerdiği besin elementleri nedeniyle önemli bir besin kaynağıdır. Bu çalışmada 173 adet fasulye genotipi kullanılarak Genotyping by Sequencing metodu ile elde edilmiş 20766 adet SNP markörü kullanılmıştır. Structure yazılımı ile yapılan analizler sonucu populasyonun 2 ana gruba ayrıldığı (K=2) saptanmıştır. Fasulye genotipleri ile danede besin elementi analizleri gerçekleştirilmiş, sonuçlar SNP markörleri ile birlkte TASSEL (Trait Analysis by Association, Evoluation and Linkage) yazılımı üzerinden yapılan Mixed Linear Model (MLM) analizleri gerçekleştirilmiştir. Analizler sonucu bakır besin elementinin danede birikimi ile yüksek ilişkili 3 adet markör tespit edilmiştir. Bu çalışma fasulyede, Cu, P, K, Mo ve Se besin elementlerinin danede birikimini, Bornova ve Menemen olmak üzere 2 lokasyon, 2015 ve 2016 olmak üzere 2 yıl toplamda 4 farklı çevre koşulu ile gerçekleştirilmiş ilk ilişki haritalama çalışmasıdır ve gelecekteki ilişki haritalama, ıslah ve markör destekli seleksiyon (MAS) çalışmalarına yardımcı olması düşünülmektedir.

Common bean is an important nutritional source in human diet due to the micronutrients it contains. In this study, 20766 SNP markers were obtained using Genotyping by Sequencing (GBS) method, from 173 common bean genotypes. Analysis that was done by Structure softwere showed that the population is split into two (K=2) main groups. Results from micronutrients analysis performed on common bean genotypes were analyzed with Mixed Linear Model (MLM) on TASSEL (Trait Analysis by Association, Evoluation and Linkage) software, along with SNP marker. According to results of these analysis, 3 markers identified that strongly associated with copper. This study is the first association mapping study about accumulation of Cu, P, K, Mo and Se micronutrients in common bean that uses four different environmental factors (two different locations as Bornova and Menemen throughout two years (2015 and 2016)), and it will help future breeding and marker assisted selection (MAS) studies.
