Abstract

Stres koşullarında, bitkilerde moleküler düzeyde gözlenen tepkilerin belirlenmesinde RNA izolasyonu ilk ve önemli aşamalardan biridir. RNA izolasyonunun yapılması ile Poly(A)+RNA, Northern Blot Hibridizasyonu, cDNA analizleri ile stres koşullarında üretilen proteinlerin özelliklerinin belirlenmesi mümkün olmaktadır. Bu çalışmalar ile bitkinin stres koşullarına adaptasyon ve tolerans mekanizması ile strese dayanıklılığı sağlayabilecek genler ortaya konabilir, yapısal özellikleri belirlenen stres proteinleri dışarıdan bitkilere uygulanarak stres koşullarına uyumlarının sağlanması ile ilgili araştırmalara zemin oluşturulabilir. Ancak tüm çalışmaların yapılabilmesi için yüksek saflıkta ve konsantrasyonda total RNA'nın izole edilmesi ön koşuldur.Bu araştırma, günümüzde giderek artan tuzluluk problemi altındaki alanlarda yetiştirilen ürünlerden olan domates bitkisinin tuz stresi altında total RNA miktarında meydana gelen değişimin ve RNA izolasyon kitlerinin domates bitkisi örneklerinde kullanım olanaklarının belirlenmesi amacıyla yapılmıştır.Deneme tam otomasyonlu topraksız tarım tekniğiyle üretim yapan sera koşullarında gerçekleştirilmiştir. İki farklı verime yatmış domates bitkisi sırasına sırasıyla 50 ve 100 mM NaCl çözeltisi, tuz stresi oluşturmak için 15 gün süre ile uygulanmış, diğer bir domates sırası ise kontrol amaçlı kullanılmıştır.Araştırma sonucunda, denemeye alınan domates bitkilerinin uç yapraklarından bitki örneği alınmış ve total RNA analizleri, RNA izolasyon kitleri ile İzmir Yüksek Teknoloji Enstitüsü Laboratuarlarında yapılmıştır. Elde edilen sonuçlar, kontrol grubuna göre 100 mM NaCl uygulanan örneklerde önemli bulunmuştur.Anahtar sözcükler: Tuz stresi, total RNA miktarı, domates

Under stress conditions, RNA isolation is the first significant step as the molecular response. By RNA isolation, Poly (A)+RNA, Northern Blot Hybridization cDNA analysis, stress protein properties would be determined. Moreover, adaptation to stress conditions, the tolerance mechanisms, stress resistant genes, application of proteins could be studied. Moreover, the prerequisite is to isolate a highly concentrated and pure total RNA.This research, today an increasing salinity problem in the area under the stress of the products that are grown tomato plants under the stress of the salt in the amount of total RNA, and the changes of the RNA isolation kit tomato plant samples to determine possible use is made of.Experiment with fully automated production soilless agricultural techniques in the greenhouse conditions were conducted. 2 different plants during tomato yield was down to 50 and 100 mm NaCl solution, respectively for a period of 15 days was applied in order to control other tomatoes were used.A result of research in the experiment received the tips of tomato plants from the leaves of the plant samples were taken and total RNA analysis, RNA isolation kits were made with the Izmir Institute of Technology laboratories. Obtained results, compared to the control group were significant in samples applied to 100 mm NaCl.Key words: Salt stress, the amount of total RNA, tomato
