Abstract

This study is about isolation, phenotypic and molecular characterization of polybetahydroxybutyrate (PHB) producing halophilic microorganisms (extremely halophilic Archaea) from İzmir Çamaltı Saltern and researching PHB yield analysis. For this aim, 14 isolates, which are isolated from saltern soils, phenotypic and genotypic analysis had done and for researching of PHB producing isolates that have biotechnological potential, they were stained with Nile Blue A as a fluorescence stain specific to PHB. Number 10 was selected as a best PHB producing isolate.In this study as a control group of PHB production, Haloferax mediterranei ATCC 33500 type strain was used. PHB yields of Number 10 and Haloferax mediterranei ATCC 33500 was analysed by using three different medium. These mediums were defined as 1% acetate and 2% glucose containing PHB medium and 2% glucose containing PHB medium with phosphate limitations and 1% glucose, 0.1 % yeast extract containing basic medium. Finally, in the %1 acetate % 2 glucose containing PHB medium with phosphate limitation, number 10 produced 6.53 % PHB, Haloferax mediterranei ATCC 33500 produced 40.3 % PHB of total dried cell mass, in the 2% glucose containing PHB medium with phosphate limitation ,number 10 produced 5.34 % PHB, Haloferax mediterranei ATCC 33500 produced 38.59 % PHB of total dried cell mass, in the 1% glucose and 0.1% yeast extract containing basic medium number 10 produced 4.48 % PHB, Haloferax mediterranei ATCC 33500 produced 54.26 % PHB of the total dried cell mass. There is a very little effect of acetate on PHB production. There was a big increase in the PHB production of Haloferax mediterranei ATCC 33500 when cultivated in basic medium but this was not suitable for number 10. As a result basic medium is best for Haloferax mediterranei ATCC 33500 but 1% acetate and 2% glucose containing medium with phosphate limitation is best for number 10. According molecular characterization number 10 is Haloferax alexandrinus with 99 % similarity.In this study we also investigated phenotypic and genotypic analysis of the 14 isolates. By sequence analyzing of 16 S r RNA genes of the 14 isolates and NCBI nucleotide blast programme; number 1, 4, 6, 7, 8, 10, 11, 13 resembles Haloferax alexandrinus with 99 %. Number 2 resembles Halobacterium salinarum R1 with 99 % similarity. Number 3 resembles Haloferax sp. HSC4 with 99 %. Number 5 resembles Halobacterum alexandrinus with 98 % similarity. Number 9 resembles Halobacterum salinarum with 97 % similarity. Number 12 resembles Halobacterum alexandrinus with 96 % similarity. By using Genebee programme phylogenetic tree of isolates was made. This study is important both researching of PHB producing extremely halophilic archaea from İzmir Çamaltı Saltern and also gives idea for biodiversity of extremely halophilic archaea in İzmir Çamaltı Saltern.Key Words: İzmir Çamaltı Saltern, Extremely Halophilic Archaea, Poly-ß-hydroxybutyrate, PHB

Bu tez çalışması İzmir Çamaltı Tuzlası'ndan PHB üretici halofilik mikroorganizmaların (ekstrem halofilik Archaea) varlığının araştırılması, identifikasyonu ve PHB verim analizi üzerinedir. Bu amaçla tuzla topraklarından izole edilen 14 izolatın fenotipik ve genotipik analizleri yapılmış ve biyoteknolojik potansiyele sahip PHB üretici izolatların araştırılması için PHB' ye spesifik fluoresans boya olarak Nile Blue A ile boyanmışlardır. En iyi PHB üretici izolat olarak 10 nolu izolat seçilmiştir.Bu çalışmada PHB üretici kontrol grubu olarak Haloferax mediterranei ATCC 33500 tip straini kullanılmıştır. 10 nolu izolatın ve Haloferax mediterranei ATCC 33500 tip straininin PHB verimleri üç değişik ortam kullanılarak araştırılmıştır. Bu ortamlar; fosfat sınırlaması olan %1 asetat ve %2 glikoz içeren PHB ortamı ve %2 glikoz içeren PHB ortamı ile % 1 glikoz, %0,1 yeast extract içeren temel ortam olarak belirlenmiştir. Sonuç olarak fosfat sınırlaması olan %1 asetat, %2 glikoz içeren PHB ortamında 10 nolu izolat toplam kuru hücre ağırlığının %6,53'ü, Haloferax mediterranei ATCC 33500 %40,3'ü; fosfat sınırlaması olan %2 glikozlu PHB ortamında 10 nolu izolat toplam kuru hücre ağırlığının % 5,34'ü, Haloferax mediterranei ATCC 33500 %38,59'u; %1 glikoz ve %0,1 yeast extract içeren temel ortamda 10 nolu izolat toplam kuru hücre ağırlığının %4,48'i, Haloferax mediterranei ATCC 33500 % 54,26'sı oranında PHB üretmiştir. Burada asetatın PHB üretimine çok az bir etkisi olduğu görülmüştür. Ayrıca temel ortamın Haloferax mediterranei ATCC 33500 için uygun fakat 10 nolu izolat için uygun olmadığı görülmüştür. Sonuç olarak % 1 glikoz, %0,1 yeast extract içeren temel ortam Haloferax mediterranei ATCC 33500 için, fosfat sınırlaması olan %1 asetat, %2 glikoz içeren PHB ortamı 10 numara için en iyi ortamdır. Moleküler karakterizasyona göre 10 nolu izolat %99 benzerlikle Haloferax alexandrinus' tur.Bu çalışma kapsamında 14 izolatın da fenotipik ve genotipik analizleri yapılmıştır.14 izolatın 16 S r RNA genlerinin dizi analizleri ve NCBI Blast programı ile 1, 4, 6, 7, 8, 10, 11, 13 nolu izolatlar Haloferax alexandrinus'a %99 benzer çıkmıştır. 2 nolu izolat %99 oranında Halobacterium salinarum R1 strainine, 3 nolu izolat %99 oranında Haloferax sp.HSC4'e, 5 nolu izolat %98 oranında H.alexandrinus'a, 9 nolu izolat %97 oranında Halobacterum salinarum'a benzemekte, 12 nolu izolat %96 oranında H.alexandrinus'a benzemektedir. Ayrıca Genebee programı kullanılarak İzolatların filogenetik ağacı oluşturulmuştur. Bu çalışma hem İzmir Çamaltı Tuzlası'ndan PHB üretici ektsrem halofilik archaea varlığının araştırılması hem de İzmir Çamaltı Tuzla'sının ekstrem halofilik Archaea biyoçeşitliliği hakkında bilgi vermesi bakımından önemlidir.Anahtar kelime: İzmir Çamaltı Tuzlası, Ekstrem Halofilik Archaea, Poli-ß-hidroksibütirat, PHB
