Abstract

vu ABSTRACT DEVELOPMENT OF AFLATOXIN Mt ANALYSIS METHOD IN KEFIR AND KEFIR GRAIN AND INVESTIGATION OF THE CARRY-OVER OF THE AFLATOXIN Mi FROM CONTAMINATED MDLK TO KEFIR AND KEFIR GRAIN KENDÎRCÎ, Perihan MSc in Food Engineering Supervisor: Prof.Dr. Tomris ALTU? September 2002, 67 Pages In this thesis, the appropriate method is developed for determination of Aflatoxin Mi in kefir and kefir grain and the carry-over of the Aflatoxin Mi from contaminated milk to kefir and kefir grain and changes that occur during storage are investigated. The effect of two bacteria isolated from kefir on Aflatoxin Mi is also observed in culture medium. As the result of the experiments conducted for analysis of Aflatoxin Mi in contaminated kefir and kefir grain, the application of immunoaffinity column with HPLC analysis technique were found as the most accurate method. The recovery values obtained by this method for kefir and kefir grain were found to be 75% and 65%, respectively. By using this method, the carry-over of Aflatoxin Mi from milk contaminated at 0.1, 0.2 and 0.5 ug/1 levels in laboratory conditions to kefir and kefir grain was investigated. The carry-over ratio of Aflatoxin Mi to kefir in respect to the contamination levels (0.1, 0.2 and 0.5 ug/1) were found to be 60, 60 and 80%, whereas they were dedected as 1.6, 2.6 and 2.6% in kefir grain. It was also determined that there wasn't any effect of storage of Aflatoxin Mi level in kefir. There weren't precise results which indicate the effect of bacteria on Aflatoxin Mi in cultured medium. ÖZET KEFİR VE KEFİR TANESİNDE AFLATOKSİN Mı TAYİN YÖNTEMİNİN GELİŞTİRİLMESİ VE KONTAMİNE SÜTLERDEN KEFİRE VE KEFİR TANELERİNE AFLATOKSİN Mı GEÇİŞİNİN ARAŞTHULMASI KENDIRCÎ, Perihan Yüksek Lisans Tezi, Gıda Mühendisliğ Bölümü Tez yöneticisi: Prof.Dr. Tomris ALTUĞ Eylül 2002, 67 Sayfa Bu tezde kefir ve kefir tanesinde Aflatoksin Mı tayini için uygun yöntem geliştirilmiş ve kontamine sütlerden kefire ve kefir tanelerine Aflatoksin Mı geçişi ve depolamanın etkisi araştırılmıştır. Ayrıca kefirden izole edilen iki bakterinin kültür ortamında Aflatoksin Mı 'e etkisi incelenmiştir. Kontamine edilmiş kefir ve kefir tanesinde Aflatoksin Mı analizi için yapılan denemeler sonucunda imünoafiniti kolon ile saflaştırmadan sonra YBSK analizi tekniğinin en doğru sonuç veren yöntem olduğu belirlenmiştir. Söz konusu yöntem kullanılarak kefir için %75, kefir tanesi için ise %65 geri kazanım değeri saptanmıştır. Aflatoksin Mı tayini için belirlenen yöntem kullanılarak laboratuvar koşullarında 0.1, 0.2 ve 0.5 ug/1 düzeylerinde Aflatoksin Mı ile kontamine edilen sütlerden kefir eldesinde bu toksinin kefire ve kefir tanesine geçişi incelenmiştir. Kefire geçiş oram sırasıyla %60, 60 ve 80; kefir tanesine geçiş oram ise sırasıyla %1.6, 2.6 ve 2.6 olarak belirlenmiştir. Depolamanın kefirdeki Aflatoksin Mı üzerine etkisinin olmadığı belirlenmiştir. Kefirden izole edilen bakterilerin kültür ortamında Aflatoksin Mı üzerine etkisi ile ilgili kesin bulgular elde edilememiştir.
