Abstract

In this study, distribution of Sialic acid (Sia) and Galactose (Gal) sugars in muscle tissue of Galleria mellonella was investigated according to body parts (head, torax, and abdomen) and developmental stages (larval stages, pupae and adult) of the insect.For this aim, first 8?m thick tissue sections were checked by hematoxylin-eosin staining and then the sections from same regions were treated with fluorescence-labeled lectins and examined under fluorescence microscope. According to signals coming from LFA (Limax flavus agglutinin) and LPA (Limulus polyphemus agglutinin) lectins that bind all Sia types; Sias were more extensively found in examined regions and stages of muscle tissues of first larval stages of G.mellonella. By the application of lectins that recognize the two or three sugar chain together with the glycosidic linkage, Sias are most extensively found in head, torax and abdomen muscles of first stage larva. Considering MAA (Maackia amurensis agglutinin) signals, Neu5Ac?2,3Galß1,4GlcNAc sugar chain is slightly reduce in head, torax and abdomen muscles of third larval stage. This reduced level is protected and it is similar in the following stages with the same staining density. According to SNA (Sambucus nigra agglutinin) signals, Neu5Ac?2,6Gal/GalNAc sugar chain starts decrease in head, torax and abdomen muscles begining from third larval stage. This decrease occurs gradually in the head muscles and suddenly in the torax muscles. Also, the decrease is similar in these two regions of pupae and young-adult individuals. The highest reduction in SNA signal density was detected in abdomen muscles while it is completely disappeared in pupae and young-adult stage. Signals from fluorescence labelled PNA (Arachis hypogaea) that recognizes Galß1,3GalNAc sugar chain were always detected in the muscle tissues of all examined regions and stages.As results; Neu5Ac?2,3Galß1,4GlcNAc chain is present in all regions and stages from larvae that just hatched to adult and its existence is gradually reduce by development. On the other hand, Neu5Ac?2,6Gal/GalNAc chain exists in young stages more extensively. By development, its signal reduces in head and torax regions and completely disaapears in abdomen region. There is no considerable difference in the distribution of Galß1,3GlcNAc sugar chain according to regions and developmental stages.

Bu çalışmada Galleria mellonella'da kas dokusunda Sialik asit (Sia) ve Galaktoz (Gal) şekerlerin bölgelere (baş, toraks, abdomen) ve gelişme evrelerine (larval devreler ile pupa ve ergin) göre dağılışları incelenmiştir.Bu amaçla, 8?m'lik doku kesitlerine hematoksilen-eosin kontrollerinden sonra flüoresan bağlı lektinler uygulanarak flüoresan mikroskopta incelenmiştir. Tüm Sia tiplerine bağlanan LFA (Limax flavus agglutinin) ve LPA (Limulus polyphemus agglutinin) lektinlerinin ışımalarına göre; G.mellonella'nın kas dokusunda ilk larval evrelerde incelenen bölgeler ve evrelerde Sia'lar daha yoğun olarak bulunmaktadır. Glikosidik bağ tipi ile ikili veya üçlü şeker dizilerini tanıyan lektinlerin uygulanmasıyla Sia'lar en yoğun 1. evre larva baş, toraks ve abdomen kaslarında bulunmaktadır. MAA (Maackia amurensis agglutinin) ışımalarına dayanarak, Neu5Ac?2,3Galß1,4GlcNAc dizisi şekerler baş, toraks ve abdomen kaslarında 3. larval evrede hafifçe azalır. Bu azalmış düzey korunarak, arkadan gelen evrelerde benzer yoğunlukta bulunurlar. SNA (Sambucus nigra agglutinin) ışımalarına göre, Neu5Ac?2,6Gal/GalNAc dizisi şekerler baş, toraks ve abdomen kaslarında 3. larval evreden başlayarak gittikçe azalır. Azalmalar baş kaslarında kademeli, toraks kaslarında birdenbire meydana gelir ve pupa ve genç erginlerin bu iki bölgesinde benzer bulunurlar. SNA ışıma yoğunluğunda en fazla azalma abdomen kaslarında görülerek pupa ve genç erginlerde tamamen kaybolur. Flüoresan işaretli PNA (Arachis hypogaea) ışımasıyla Galß1,3GalNAc dizisi şekerler incelenen bütün bölgelerde ve devrelerdeki kas dokularında kararlı şekilde daima bulunur.Sonuç olarak; Neu5Ac?2,3Galß1,4GlcNAc dizisi yumurtadan yeni çıkmış larvadan ergine kadar tüm bölge ve devrelerde, gelişme ilerledikçe azalarak bulunmaktadır. Neu5Ac?2,6Gal/GalNAc dizisi ise genç evrelerde daha yoğun bulunur. Gelişme ilerledikçe baş ve toraks bölgelerinde azalırken abdomen bölgesinde tamamen kaybolur. Galß1,3GlcNAc dizisi şekerlerin dağılışında bölgelere ve gelişme evrelerine göre önemli fark bulunmamaktadır.
