Abstract

The main objective of this study was to optimize gerbera propagation throughtissue culture to produce good quality seedling. Flower buds (1.0-1.5 cm diameter)showed the best shoot regeneration compared to medium sized or fully mature ones.The shoot tip showed the best shoot regeneration in a medium containing BA at 5.0mg/l . Only flower stems of the cultivar Red Bull formed shoots . Shoots formed on invitro leaves in media with both low or high BA concentration but not in BA-freemedium . Multiplication rate increased with increasing BA concentration in themedium. Multiplication and vitrification rates decreased with the increase of theconcentration of the different gelling agents in the medium . All the rooting mediatested gave 100 % rooting. After transfer from the agar medium to in vivoconditions, plantlets showed a survival percentage of more than 90 % in bothcocopeat alone or cocopeat + perlite at the ratio of 1:1.

Bu projenin ana amacı, gerberanın doku kültürü yoluyla çoğaltımınınoptimizasyonu ile kaliteli fide elde edilmesidir.1.0-1.5 cm çapındaki çiçek tomurcukları,daha büyük tomurcuklara ve tam olgunlaşmış çiçeklere göre en iyi sonucu vermiştir.Sürgün ucu eksplantları en iyi sonucu 5 mg/l BA içeren ortamda göstermiştir. Çiçek sapıeksplantları yalnızca Red Bull varyetesinde sürgün oluşturmuştur. Yüksek veya düşükBA konsantrasyonlarında in vitro yapraktan sürgün oluşumu gözlenmiş ancak BAiçermeyen ortamda sürgün oluşmamıştır. Ortamdaki BA konsantrasyonu ile kardeşlenmedoğru orantılıdır. Jel yapıcı maddelerin ortamdaki konsantrasyonlarının artması ilevitrifikasyon ve kardeşlenme oranında azalma saptanmıştır. Denenen tüm köklenmeortamlarında % 100 başarı sağlanmıştır. İn vitro'dan in vivo'ya transfer edilenbitkiciklerde cocopeat veya cocopeat-perlit (1:1) ortamlarında % 90'dan fazla tutumgözlenmiştir.
