In vitro formation of gerbera (Gerbera jamesonii Bolus) plantlets from capitulum explants

Abstract

The shoot regeneration capacity of gerbera capitulum explants at different ages was investigated. The effect of the exogenous cytokinin BA concentration on in vitro shoot regeneration on the capitulum explant was studied. Regenerated shoots were separated from the capitulum explants and cultured in nutrient media containing cytokinins BA or kinetin for multiplication. For in vitro rooting shoots were then transferred to rooting media with different concentrations of nutrient elements, IBA and IAA. In vitro rooted shoots were transferred to in vivo conditions and planted in cocopeat or in a mixture of cocopeat + perlite (1:1) for acclimatization. Immature capitula of 1.0-1.5 cm in diameter showed the best shoot regeneration capacity compared to those at medium age or fully mature ones. The gerbera cultivars tested showed different responses to cytokinin (BA) concentration with regard to shoot formation on the capitulum explants. The multiplication rate increased with increasing BA concentration in the medium. The medium containing BA at 2.5 or 5.0 mg l-1 gave a higher multiplication rate than the medium with kinetin at 10.0 mg l -1. All the rooting media tested gave 100% rooting. There were very slight differences between the different rooting media with respect to number of roots per shoot and root length. Results showed no significant difference in rooting between full or half strength Murashighe and Skoog (1962) medium. After transfer from the agar medium to in vivo conditions, plantlets showed a survival percentage of more than 90% in either cocopeat alone or cocopeat + perlite at the ratio of 1:1.
