Abstract

ÖZET NİŞİN ÜRETEN L.lactis İZOLASYONU VE NİŞİN ÜRETİMİNE ETKİ EDEN FAKTÖRLER KIŞLA, Doygu Doktora Tezi, Gıda Mühendisliği Bolümü Tez Yöneticisi : Prof. Dr. Adnan ÜNLÜTÜRK Nisan 2001, 133 sayfa Bu çalışmada, agar kuyu difüzyon (agar well-diffusion method) ve nokta inokülasyon agar difüzyon (agar spot method) yöntemleri ile nişin üreten L.lactis izole edilmeye çalışılmıştır. Toplam 83 örneğin 67'sinden izole edilen 273 laktik asit bakterisi bakteriyosin üretimi açısından taranmış ve bakteriyosin üreten bir izolata rastlanmamıştır. Nişin üreten L.lactis 497 suşunun nisin aktivitesi hem regresyon eşitliği kullanılarak (IU/ml) hem de nisin titresi (AU) olarak saptanmaya çalışılmıştır. Her iki yöntemin de nişin aktivitesinin belirlenmesinde sağlıklı ve hassas yöntem olmadıkları bulunmuştur. Regresyon eşitliği kullanılarak saptanan nişin aktivite değerleri daha fazla değişken sonuçlar verdiği için, nişin üretimine etki eden faktörlerin (karbon kaynaklarının çeşidi ve konsantrasyonu, maya ekstraktı konsantrasyonu, mangan iyonu, magnezyum iyonu, kalsiyum iyonu konsantrasyonları) belirlenmesinde, nişin aktivitesi nişin titresi olarak hesaplanmıştır. Elde edilen sonuçlar sakkarozun diğer karbon kaynaklarına (glukoz, fruktoz, laktoz) kıyasla nişin aktivitesini artırdığı, sakkaroz ve maya ekstraktı konsantrasyonlarının artırılmasıylaVI nişin aktivitesinde değişiklik olmadığı saptanmıştır. Aynı şekilde ortamda bulunan mangan, magnezyum ve kalsiyum iyonlarının nişin üretimine bir etkisinin olmadığı belirlenmiştir. Anahtar sözcükler Nişin, L.lactis, nişin aktivite tayini

Supervisor: Prof Dr. Adnan UNXUTURK April 2001, 133 pages Isolation of nisin producing LAactis by using agar well- diffusion and agar spot methods has been studied Totally 273 lactic acid bacteria isolated from milk, cheese and vegetable samples were screened for bacteriocin production by using indicator microorganism M.luteus 982 and no bacteriocin producer was detected. Determination of the nisin activity of Llactis 497 by regression equation (IU/ml) and as a nisin titer has been studied. Both of the methods were found as not sensitive and reliable. Since nisin activity values determined by regression equation gave more unreliable results as compared to the nisin titer method, as the activity indicator of nisin, nisin titer has been used in the determination of the effect of various factors (type and concentration of carbon sources, yeast extract concentration, concentrations of manganese, magnesium and calcium ions) in nisin production of LAactis 497. Sucrose, in comparison with other carbon sources (glucose, fructose, lactose), increased the nisin activity and increasing the sucrose and yeast extract concentrations did not cause any increase in nisin production. Similarly, ions suchvııı as manganese, magnesium and calcium did not have any effect on nisin production. Key words: Nisin, Llactis, nisin activity determination
