Abstract

The cervids, as well as other free-living brazilian species, have suffered pressures on their habitat, causing segregation between populations and reduced genetic exchange, which may decrease the genetic diversity and generate increased inbreeding and extinction risk. In vitro embryo production (IVEP) has been highlighted as a tool to be used in wild species conservation programs in in situ and ex situ. The maturation process has a great importance among the steps of IVEP, and its success can ensure an improvement in the rate of production of embryos. The objectives of this study were to assess feasibility of the protocol of ovarian stimulation for ovum pick, the efficiency of laparoscopy for oocyte retrieval, and the nuclear maturation and mitochondrial distribution in oocytes matured in vitro for the species M. gouazoubira. Thus, we used five females, submitted to repeated stimulation with intervals of 30 days. After the visualization of the ovaries, the visible follicles were counted and punctured by laparoscopy on day eight (D8). The aspirated oocytes were classified morphologically according the quality, subjected to in vitro maturation for 24 hours and then stained for the evaluation of mitochondrial distribution (Mitotracker Red CMXRos) and nuclear maturation stage (Hoescht 33342). Fourteen surgical procedures were performed on overstimulated females and 94 oocytes were collected in total. The average number of visualized follicles, aspirated follicles, and harvested oocytes was equal to 17.07±9.12, 13.54±6.64 e 7.15±3.72, respectively. Of the in vitro matured oocytes, 64.52% reached the MII stage and 67.74% were classified with homogeneous mitochondrial distribution. Assim como outras espécies brasileiras de vida livre, os cervídeos têm sofrido pressões sobre seu habitat, o que provoca segregação entre populações, diminuição de troca gênica e de diversidade genética, aumento da endogamia, e um maior risco de extinção. A produção in vitro de embriões (PIVE) tem ganhado destaque como uma ferramenta a ser utilizada nos programas de conservação de espécies selvagens in situ e ex situ. Entre as etapas da PIVE, a maturação in vitro (MIV) assume grande importância, pois o seu sucesso pode garantir a melhora nas taxas de produção de embriões. Assim, os objetivos do presente trabalho foram: avaliar viabilidade do protocolo de estimulação ovariana para a aspiração folicular, a eficiência da laparoscopia para a aspiração folicular, e a maturação nuclear e a distribuição mitocondrial em oócitos maturados in vitro para a espécie M. gouazoubira. Para isso, foram utilizadas cinco fêmeas, submetidas a repetidas estimulações ovarianas com intervalos de 30 dias. Os oócitos aspirados foram classificados morfologicamente de acordo com a qualidade, submetidos à maturação in vitro por 24 horas e, posteriormente, corados para a avaliação da distribuição mitocondrial (Mitotracker Red CMXRos); e do estágio de maturação nuclear (Hoescht 33342). Foram realizados 14 procedimentos cirúrgicos para a colheita de 94 oócitos de fêmeas superestimuladas. A média de folículos visualizados, folículos aspirados e oócitos colhidos por cirurgia foi, respectivamente, de 17,07±9,12, 13,54±6,64 e 7,15±3,72.
