Isolamento de linhagens fúngicas termofílicas produtoras de pectinases termoestáves: produção, caracterização e purificação parcial da poligalacturonase

Abstract

Fungi, due to their reproduction and growth characteristics, are well adapted to a large variety of substrates, being excellent decomposers of vegetal matter. Pectinases produced by thermophilic fungi exhibit important characteristics for application in bioprocesses, such as stability towards pH and temperature. The type of fermentation process used for the production of these enzymes has great influence on the amount of enzyme that is produced and on their properties. The goal of this work was to isolate and identify thermophilic fungal strains that produce pectinases and cultivate them in solid state fermentation (SSF) and in submerged fermentation (SmF) in order to evaluate their potential to produce polygalacturonases (PG), as well as to characterize the PG produced. Samples (0.5 g) were collected from piles of composting material and from deposits of agricultural and agro-industrial residues, which were transferred to tubes containing pectin as the only carbon source. The isolated fungal strains were cultivated at 45°C, for 120 hours in SSF, using wheat bran (WB), orange peel (OP), sugar cane bagasse (SB) and washed orange bagasse (WP) as substrates and in SmF, using a liquid nutrient medium containing 1.0% pectin as carbon source. Samples were taken every 24 hours and the enzymatic activity was assayed through the quantification of galacturonic acid released by citrus pectin (1.0%) hydrolysis. Of a total of 40 samples collected, 34 fungal strains were isolated, of which the best PG producers in SmF were Thermomyces sp N4 (1.8 U/mL) and Scopulariopsis sp N7.1 (0.74 U/mL). The best PG producers in SSF were Thermomucor sp N31 (15.0 U/g), Aspergillus sp N12 (76.1 U/g) and Aspergillus sp N29 (64.7 U/g). Os fungos, em função de suas características de reprodução e crescimento, adaptam-se a uma grande variedade de substratos, sendo excelentes decompositores de material orgânico. As pectinases produzidas por fungos termofílicos apresentam características importantes para aplicação em bioprocessos, como estabilidade ao pH e à temperatura. O tipo de processo fermentativo utilizado para a obtenção dessas enzimas influência a quantidade obtida e as propriedades das mesmas. O presente trabalho teve como objetivos isolar e identificar linhagens fúngicas termofílicas produtoras de pectinases e cultivá-las por FES e por FSM para a avaliação do potencial de produção de poligalacturonases, além de caracterizar a PG produzida. Foram coletadas amostras (0,5 g) de compostagem e depósitos de resíduos agrícolas e agro-industriais, as quais foram transferidas para tubos contendo pectina como única fonte de carbono. As linhagens fúngicas isoladas foram cultivadas a 45°C, por 120 horas em FES, utilizando-se como substratos, farelo de trigo (FT), bagaço de laranja industrializado (BL), bagaço de canade- açúcar (BC) e casca de laranja lavada (CL) e em FSM utilizando-se meio nutriente líquido contendo 1% de pectina como fonte de carbono. As amostras foram retiradas a cada 24 horas e a atividade enzimática foi avaliada a partir da quantificação do ácido galacturônico liberado por hidrólise de pectina citrus (1%). De um total de 40 amostras coletadas, foram isoladas 34 linhagens fúngicas entre as quais as maiores produtoras de poligalacturonases por FSM foram Thermomyces sp N4 (1,8 U/mL) e Scopulariopsis sp N7.1 (0,74 U/mL). As maiores produtoras de PG em FES foram as linhagens Thermomucor sp N31 (15 U/g), Aspergillus sp N12 (76,1 U/g) e Aspergillus sp N29 (64,7 U/g).
