Abstract

O linfoma é uma doença linfoproliferativa caracterizada pela multiplicação clonal de linfócitos malignos no seio dos órgãos linfóides, afetando principalmente os linfonodos, ou outros órgãos viscerais sólidos, com prognóstico dependente de vários fatores, incluindo invasão tecidual. Enzimas proteolíticas MMP-2 e MMP-9 estão sendo estudadas quanto à importância neste processo de progressão tumoral por propiciarem a penetração e infiltração celular. Considerando a importância atual destas enzimas e o papel ainda não fundamentado nas neoplasias hematopoiéticas, avaliou-se a expressão de MMP-2 e MMP-9 em cães acometidos por linfoma multicêntrico e cães hígidos, através de técnica imuno-histoquimica de linfonodo e eletroforese zimográfica de linfonodo e soro, estabelecendo a relação entre a expressão destas enzimas e os fatores prognósticos como estadiamento, imunofenotipo e grau histopatológico da neoplasia e resposta ao tratamento. Observou-se maior expressão tecidual de MMP-9 nos animais com linfoma, através de técnica imuno-histoquimica, sem correlação exata com prognóstico. Eletroforese zimográfica do linfonodo demonstrou correlação da MMP-9 com imunofenótipo T e animais em doença progressiva, inferindo possível prognóstico negativo quanto à sobrevida. Valores de MMP-2 não apresentaram relação clara com o processo tumoral, assim como os valores séricos das duas enzimas. Desta forma, a MMP-9 demonstrou participação no processo neoplásico, porém sem determinação prognóstica exata

Lymphoma is a lymphoproliferative disease characterized by clonal proliferation of malignant lymphocytes within lymphoid organs, mainly affecting the lymph nodes, or other solid visceral organs, with prognosis depends on several factors, including tissue invasion. Proteolytic enzymes MMP-2 and MMP-9 have been studied as to the importance of tumor progression in this process by propitiate the penetration and cellular infiltration. Considering the current importance of these enzymes and the role not grounded in hematopoietic malignancies, we evaluated the expression of MMP-2 and MMP-9 in dogs affected by multicentric lymphoma and healthy dogs by immunohistochemistry technique of lymph node and zymography electrophoresis of lymph node and serum, establishing the relationship between the expression of these enzymes and prognostic factors such as tumor staging, immunophenotyping, histological grade and response to treatment. We observed increased expression of MMP-9 tissue in animals with lymphoma by immunohistochemistry technique, no exact correlation with prognosis. Zymography electrophoresis of lymph node demonstrated correlation of MMP-9 with T-cell phenotype and progressive disease, inferring possible negative prognosis regarding survival. Values of MMP-2 showed no clear relation with the tumoral process as well as serum levels of the two enzymes. Thus, MMP-9 demonstrated involvement in the neoplastic process, but without accurate prognostic determination
