Abstract

In aerobic living organisms, physiologic processes produce reactive oxygen species (ROS) as by-products or to fight against microorganisms. This species have being studied, whereas promotes DNA mutation, aging process and several pathologies. Reactive species and free radicals generation are equilibrated by antioxidants. Thus, these antioxidants in diets, are protector agent to the human body in removal of free radicals. Since antiquity the mushrooms are consumed as a high nutritional meal and therapeutic worth. In this manner, the search of new medicines turns natural products research an important option for discovering molecules with different biological activities. Natural products research is relevant for discovering molecules able to fight against oxidant species. In evaluating the antioxidant potential of substances, it is important to considerate: a compound should be tested at concentrations achievable in vivo and in assaying antioxidants, one should use biologically relevant ROS. It is important to know if it works directly over the oxidant or if it works by regulating endogenous antioxidants defenses. Thus, this work had as objective the partition (using chromatografic techniques, as: TLC and HPLC) and validation, in vitro, of the Agaricus blazei Murril mushroom fractions as antioxidant products (using préestablished methodologies, as: ABTS radical, DPPH radical, Superoxide radical, TNB/HOCl and TNB/H2O2). It was also evaluated, the presence of peroxidase and polyphenoloxidase enzymes. The evaluated fractions showed antioxidant activity similar to the patterns (trolox and uric acid) in removal of ABTS radical and DPPH radical, except for the hexanic fraction. In the TNB/HOCl system, the samples presented small activity (it wasn't possible to determi ne IC50), and your activities were very inferior to the patterns. In the TNB/H2O2 system, the samples didn't presented activity to kidnap H2O2.

Nos organismos vivos aeróbios, processos fisiológicos produzem espécies reativas de oxigênio (ERO) como subprodutos ou com a finalidade de combater microorganismos invasores. Estas espécies têm sido estudadas, pois promovem mutações de DNA, processos de envelhecimento e várias patologias. A formação de espécies reativas e radicais livres são equilibradas naturalmente pela existência de compostos conhecidos como antioxidantes. Tais compostos na dieta, são agentes protetores do corpo humano na remoção dos radicais livres. Os cogumelos são fungos conhecidos, desde a antiguidade, devido sua utilização, pelo homem, como alimento de elevado valor nutritivo e terapêutico. Neste sentido, a busca por opções terapêuticas para diferentes patologias faz da pesquisa de produtos naturais um campo fértil em opções de moléculas com diferentes atividades biológicas. A relevância da pesquisa de produtos naturais proporciona a descoberta de novos fármacos e o estudo de cogumelos que apresentem substâncias que possam agir sobre as diferentes espécies oxidantes geradas em nosso organismo torna-se de grande importância. No estudo da avaliação do potencial antioxidante das amostras deve-se considerar que: um composto deve ser testado em concentrações disponíveis in vivo e ao avaliar os antioxidantes, deve-se utilizar pelo menos uma espécie relevante biologicamente. Deve-se perguntar como age o antioxidante, se ele age diretamente sobre a ERO ou ele inibe a sua geração e, ainda, se ele age indiretamente regulando defesas antioxidantes endógenas. Assim, este trabalho teve por objetivo a partição (usando técnicas cromatográficas como: CCD e CLAE) e validação, in vitro, das frações do cogumelo Agaricus blazei Murril como produtos antioxidantes (usando metodologias préestabelecidas, como: radical ABTS, radical DPPH, Radical Superóxido, TNB/HOCl e TNB/H2O2).
