The aim of this study was to evaluate the pupal response to different adhesives systems directly applied on mechanically exposed pulps in rats. The oclusal surfaces of first and second molars were prepared and the pulps exposed. In C (control group-n=34) pulps were capped with a calcium hydroxide paste (Dycal); E1 (n=31) received the Single Bond adhesive system and E2 (n=30) was treated with the self-etching One-up Bond F adhesive system. The materials were applied according to the manufacture's instructions and cavities were restored with Z100 composite resin. Animals were sacrificed after seven, thirty and 45 days and histological features of the dental pulp were evaluated. The Fisher test was used as statistical analysis. At seven days C and E2 presented similar inflammatory response and pulpal organization, showing focal chronic inflammation and localized odontoblast cell lost. E1 and E2 demonstrated statistically significant difference regarding to pulpal organization and inflammatory response; the former exhibiting much more necrosis. At seven days, bacterial contamination of the dental pulp was less evident with One-up Bond F. All these differences were lost with time and after 45 days, pulpal necrosis and bacterial contamination prevailed in the three groups. The formation of the dentin barrier was greater with time in the three groups. The results presented suggest negative effects of direct pulp capping on molar teeth of rats using Dycal, One-up Bond F and Single Bond, under the experimental conditions described.
