Atividade antibacteriana de óleos essenciais e avaliação de potencial sinergético

Abstract

Research about medicinal properties of plants is focused on several areas, and its antimicrobial activities and potential use against multiresistant strains have received special attention. This study aimed to assess the antimicrobial potential of seven essential oils (EO) alone, and the possible synergism of some of these EO when combined with each other and with standard antimicrobials against clinically important bacterial strains with different resistance profiles, among them, Staphylococcus aureus (methicillin-resistant (MRSA) and methicillin-sensitive (MSSA) STRAINS), Escherichia coli, Pseudomonas aeruginosa (all human clinical isolates), and Salmonella spp. (clinical and food samples). The tests were divided into two steps. Initially (Article I), we obtained the minimum inhibitory concentration (MIC) for assa peixe (Vernonia polyanthes L.), rosemary (Rosmarinus officinalis L.), alecrim do campo (Baccharis dracunculifolia DC), clove (Caryophyllus aromaticus L.), cinnamon (Cinnamomum zeyalnicum L.), chamomile (Matricaria recutita L.) and Brazilian cherry (Eugenia uniflora L.) oils, by agar dilution method. The OE were characterized by gas chromatography-mass spectrometry (GC-MS) and the chemical composition of each oil was related to the antimicrobial activity against the different bacterial profiles. In the second part of the study (Article II), based on the action potential and yield of extraction of the oils, we evaluated the synergism between the four oils with higher potential, as well as the synergism with tetracycline and oxacillin against S. aureus, E. coli and P. aeruginosa. The combinations between EO and antibiotics were tested by the disc diffusion method. The synergistic potential of the oils was also evaluated by this method and by the microdilution. As pesquisas sobre as propriedades medicinais de plantas estão focadas em diversas áreas, tendo a atividade antimicrobiana e o uso potencial contra linhagens multirresistentes recebido atenção especial. O presente trabalho teve como objetivo geral avaliar o potencial antimicrobiano de sete óleos essenciais (OE) isoladamente, e verificar o possível sinergismo de alguns destes OE quando combinados entre si e com antimicrobianos padronizados sobre linhagens bacterianas de importância clínica com perfis de resistência diferenciados, sendo estas de Staphylococcus aureus (linhagens meticilina-resistentes, MRSA, e meticilina-sensível, MSSA), Escherichia coli, Pseudomonas aeruginosa (todos isolados clínicos humanos) e Salmonella spp. (isolados clínicos e de alimentos). Os ensaios foram divididos em duas etapas. Inicialmente (Artigo I), foi obtida a Concentração Inibitória Mínima (CIM) para os óleos de assa-peixe (Vernonia polyanthes L.), alecrim (Rosmarinus officinalis L.), alecrim do campo (Baccharis dracunculifolia D.C.), cravo da Índia (Caryophyllus aromaticus L.), canela (Cinnamomum zeyalnicum L.),camomila (Matricaria recutita L.) e pitanga (Eugenia uniflora L.), através da metodologia de diluição em Agar. Os OE foram caracterizados através de cromatografia gasosa acoplada a espectrometria de massas (CG-EM), sendo a composição química de cada óleo relacionada com sua atividade sobre os diferentes perfis bacterianos. Na segunda parte do estudo (Artigo II), baseando-se no potencial de ação e rendimento de extração dos óleos, foi avaliado o sinergismo entre os quatro óleos com maiores potenciais, bem como a atividade conjunta destes com tetraciclina e oxacilina frente S. aureus, E. coli e P. aeruginosa.
