Annually, invasive fungal infections (IFIs) are responsible for the death of approximately 1.5 – 2 million patients. Current treatment with antifungals is limited and the increase in drug resistance demands the search for new therapeutic agents. The enzyme Gwt1, involved in the biosynthesis of glycosylphosphatidylinositols (GPIs) has acyl-transferase-like activity and is involved in the acylation of inositol in the initial step of the biochemical pathway, and proves to be a promising target in the search for new antifungals. In this work, using the molecular modification strategy called bioisosterism, seven compounds based on manogepix (APX001A), known to be a Gwt1 inhibitor, were designed. To obtain these compounds, convergent synthesis was used in seven steps. In step A, 6-carboxybenzo[c][1,2,5]oxadiazole 1-oxide (8) and 6-carboxy-[1,2,5] benzofuroxanes were obtained. ]oxadiazole[3,4-b]pyridine 1-oxide (9); in step B, the ethers N-(3-(phenoxymethyl)phenyl)-acetamide (13) and N-(3-(pridin-2-oxy)methyl)phenyl)-acetamide (14) were obtained through a reaction of bimolecular nucleophilic substitution followed by a basic hydrolysis to obtain the compounds N-(3-(phenoxymethyl)aniline (15) and N-(3-(pridin-2-oxy)methyl)aniline (16) , in step C, the coupling reaction between the compounds obtained in Blocks A and B to obtain the benzofuroxane derivatives (17) and (18); in step D, the sulfonated intermediate (22) was obtained through a bimolecular nucleophilic substitution reaction; in step E there was a coupling reaction between the sulfonated derivative (22) and the compounds obtained in Block A to obtain compounds (23) and (24); in step F there was a reduction reaction of a nitroaromatic group (25) to an aromatic amine (26) The intermediates and final products obtained in this work had their yields ranging from 21% to 89%. Structural analysis was performed using analytical methods such as one-dimensional nuclear magnetic resonance (NMR), infrared spectroscopy and fusion range. The new compounds (17, 18, 23, 24, 27, 28 and 30) designed as antifungals may constitute a new alternative for the treatment of invasive fungal infections.
