Abstract

A busca pela diminuição dos danos causados pelo envelhecimento extrínseco, ou seja, aquele causado por agentes externos, como a radiação ultravioleta, tem sido alvo de estudo. Atualmente, consumidores buscam produtos que, além da função antienvelhecimento, sejam multifuncionais e que supram todas as suas necessidades. Novas moléculas, ativos, misturas e sinergias têm buscado satisfazer um mercado cada dia mais informado. Não só a redução do envelhecimento precoce, mas, também, a solução deste problema com o uso de ativos considerados naturais e pouco agressivos. Além disso, é sabido que a busca pela segurança e eficácia deixou de ser um luxo, para se tornar parte fundamental e conduzida por leis, como a RDC 07 de 2015, por exemplo. A jabuticaba possui estudos amplos na área alimentícia, e vem sendo estudada devido seu elevado valor nutritivo e composição rica, em cosmetologia. Sua aplicabilidade tem se mostrado assertiva. Este trabalho tem o objetivo de avaliar a segurança e eficácia in vitro do uso do extrato da casca da jabuticaba, que poderá ser indicada como um novo ativo antienvelhecimento, antisséptico e multifuncional. Para isso, o trabalho foi realizado em fases: obtenção do extrato das cascas da jabuticaba e sua avaliação em parâmetros como solubilidade, varredura espectrofotométrica e quantificação de biomarcadores. Depois disso, foram avaliados o potencial de irritação cutânea e citotoxicidade in vitro (potencial de irritação epidérmica, dérmica e hepatotoxicidade). Assim, foi obtida uma faixa considerada segura de uso do extrato, sem causar danos às células da pele e fígado, caso os ativos do extrato penetre a barreira cutânea e adentrem na corrente sanguínea. Com valores seguros estabelecidos, foi avaliado o potencial de atividade antimicrobiana em microrganismos comensais e patogênicos, além da capacidade de indução da produção de colágeno e cicatrização em células HDFa. O extrato da casca da jabuticaba mostrou-se muito próspero para uso em fitocosméticos, atuando com vários efeitos benéficos, como elevada atividade antioxidante e elevada concentração de antocianinas, citotoxicidade com IC50 superiores a 6,0 mg/mL, capacidade antimicrobiana em todas as cepas testadas, além de efeito de indução na produção de colágeno in vitro.

The search for the reduction of damage caused by extrinsic aging, that is, this phenomenon by external agents, such as ultraviolet radiation, has been the subject of study. Currently, consumers are looking for products that, in addition to the antiaging function, are multifunctional and meet all their needs. New molecules, assets, mixtures and synergies have sought to satisfy a market more and more. Not only the reduction of premature aging, but also the solution of this problem with the use of assets considered natural and not very aggressive. In addition, it is known that the search for safety and efficacy provided to be a luxury, to become a fundamental part and guided by laws, such as RDC 07 of 2015, for example. Jabuticaba has extensive studies in the food area, and has been studied due to its high nutritional value and rich composition, in cosmetology. Its applicability has to be declared assertive. This work has the objective of evaluating an in vitro safety and efficacy of the use of jabuticaba bark extract, which can be chosen as a new anti-aging, antiseptic and multifunctional active. For this, the work was carried out in phases: obtaining the extract of the jabuticaba bark and its evaluation in parameters such as solubility, spectrophotometric scanning and quantification of biomarkers. After that, the skin irritation potential and in vitro cytotoxicity (potential for epidermal, dermal irritation and hepatotoxicity) were taken. Thus, it was obtained a band considered safe to use the extract, without causing damage to the skin and liver cells, if the extract's assets penetrate the skin barrier and enter the bloodstream. With safe values, the potential for antimicrobial activity in commensal and pathogenic microorganisms was evaluated, in addition to the ability to induce collagen production and healing in HDFa cells. The extract of the bark of the jabuticaba proved to be very successful for use in phytocosmetics, acting with several beneficial effects, such as high antioxidant activity and high concentration of anthocyanins, cytotoxicity with IC50 above 6.0 mg / mL, antimicrobial capacity in all strains tested, in addition to the induction effect on collagen production in vitro.
