Abstract

The damages in human health caused bytheprolonged exposure to emerging contaminants, such as hormones, is related to theaction of these compounds at the molecular level on the plasma membrane. Therefore, this work investigated the effect (causes and consequences) of the hormone 17 α-ethinylestradiol (EE2) in mono and lipid bilayers, composed of Langmuir films and giant unilamellar vesicles(GUVs), respectively. Such systems were formed using binary and ternary mixtures of the components that represent the main lipid groups of cells: 1,2-dipalmitoyl-sn-glycerol-3-phosphocholine (DPPC), cholesterol (Chol), sphingomyelin (SM) and 1-palmitoil -2-oleoyl-sn-glycero-3-phosphatidylcholine (POPC). The π-A isotherms showed that EE2interacts at the liquid/air interface with all monolayer compositions, with the higher effect on binary mixtures containing 30% Chol (DPPC/Chol and SM/Chol) and forbothneat monolayer ofPOPCand SM. In the case of the ternaries of DPPC/Chol/SM, athigh surface pressures, the presence of the hormone caused the displacement of the π-A isothermto smaller areas. The PM-IRRAS measurements indicated that the electrostatic interactions of EE2 with groups of the polarhead of DPPC and SM were kept in the binaryand ternarymixtures, causing changes in the ordering of the tails of such lipids in the mixed monolayers. The bands referring to the POPC tail region underwentthe major changes in the presence of EE2, indicatingthe presenceof the hormone in the hydrophobic phospholipid chains. In GUVs, EE2 caused changesin the spherical shape of the neat DPPC vesicles and the loss of phase contrast in the vesicles composed of DPPC/Chol and DPPC/SM. These two effects were verified simultaneously in the GUVs composed bySM/Chol. Other effects such as rupture of vesicles and formation of new structures (buds and lipidprotrusion) were verified for the DPPC/SM/Chol 1/1/1 and POPC GUVs, respectively. This work bringsan understanding of the interfacial behavior of the hormone EE2 on lipid structures and allows correlationswith biological processes that take placein-vitroandin-vivo.

Os danos causados àsaúde humana pela exposição prolongadaacontaminantesemergentes, como hormônios, são ligados à ação em nível molecular desses compostos na membrana plasmática. Portanto, este trabalhoinvestigouo efeito(causas e consequências)do hormônio 17 α-etinilestradiol (EE2) em mono-e bi-camadas lipídicas:filmes de Langmuir e vesículas unilamelares gigantes(GUVs), respectivamente. Tais sistemas são formados por misturas binárias e ternárias de componentesque representam os principais grupos lipídicos das células: 1,2-dipalmitoil-sn-glicerol-3-fosfocolina (DPPC), colesterol (Chol), esfingomielina (SM) e 1-palmitoil-2-oleoil-sn-glicero-3-fosfatidilocolina (POPC).As isotermas π-Amostraram que o EE2 interage na interface líquido/ar comtodas ascomposições de monocamadas, com maior efeitoparaas bináriascontendo 30%de Chol(DPPC/Chol e SM/Chol) e paraasmonocamadaspurasde POPCe SM.No caso dasternárias deDPPC/Chol/SM,em regiões de altas pressões superficiais, a presença do hormônio causouo deslocamento da isoterma π-A para menores áreas.As medidas de PM-IRRASindicaram que asinteraçõeseletrostáticasdoEE2 com grupos polares da cabeçadoDPPC e SMforam mantidas nas misturas bináriase ternárias, causando mudanças no ordenamento das caudas de tais lipídios nestas monocamadas mistas. As bandas referentes à região da cauda do POPC sofreram grandes mudanças na presença do EE2, indicandoa presença do hormônio nas cadeias hidrofóbicas do fosfolipídio. Nas GUVs,o EE2 causou umaalteração noformato esférico das vesículas de DPPC e a perda de contraste de fase nas vesículas compostas por DPPC/Chol e DPPC/SM. Estes dois efeitos foram verificados simultaneamente nas GUVs compostas por SM/Chol.Efeitos como ruptura das vesículas e formação de novas estruturas (buds e cordões lipídicos) foram verificados para as GUVs de DPPC/SM/Chol 1/1/1 e POPC, respectivamente. Este trabalho contribui para uma melhor compreensãodos efeitos interfaciaisdo hormônio EE2sobre estruturas lipídicase permite umacorrelaçãocom os processos biológicos que ocorrem in-vitro ou in-vivo.
