Eficácia do levamisol como imunoestimulante, redutor de estresse e antiparasitário no pacu

Abstract

The present study analyzed the levamisole hydrochloride (CL) action in the stress and innate immunity pacu (Piaractus mesopotamicus). Fish (180 ± 0.27 g; 16 ± 0.4 cm) were fed a diet containing CL for fifteen days. A control group and four treatments (T) were tested: T1 (100), T2 (150), T3 (300) and T4 (500) mg kg-1 CL, 15 fish/repeat, in quadruplicate. After that, fish (n = 40) were submitted to air exposure for three minutes, inoculated with the bacteria Aeromonas hydrophila and sampled one, 3, 6 and 24 h after inoculation. Were determined stress indicators (plasma cortisol, glucose and red blood cell count) and immunological indicators (leukocytes respiratory activity, complement system - ACH50 - and serum lysozyme). CL significantly reduced the plasma concentration of cortisol in one (500 mg kg-1) and 3 h (300 mg kg-1) after bacterial inoculation, compared with control fish. CL did not influence blood glucose, the number of erythrocytes and the serum concentration of lysozyme. CL (100 mg kg-1) increased the ARL in 1h and also the ACH50 in 3 h and all treatments 24 h after inoculation. The results of this study indicate that air exposure and bacterial infection were stressful for pacu, causing increased blood glucose and prejudice of lysozyme immune response and the CL was efficient to reduce cortisol and increased the phagocytic activity of macrophages and ACH50 in fish. During the initial sampling to analyze the effectiveness of levamisole hydrochloride as antiparasitic, four fish from control group and each treatment (n = 20) were euthanized by medullary section and sent for parasitological analysis. To analyze the histological lesions ten fish from control group and each treatment (n = 50) were euthanized in benzocaine (100 mg L-1). o presente estudo analisou a ação do cloridrato de levamisol (CL) no estresse e imunidade inata de pacu (Piaractus mesopotamicus). Os peixes (180 ± 0,27g; 16 ± 0,4cm) foram alimentados com ração contendo CL durante quinze dias. Um grupo controle e quatro tratamentos (T) foram testados: T1 (100), T2 (150), T3 (300) e T4 (500) mg kg-1 de CL, 15 peixes/repetição, em quadruplicata. Após isso, os peixes (n=40) foram submetidos à exposição aérea e inoculados com bactéria Aeromonas hydrophila e amostrados uma, 3, 6 e 24h após a inoculação. Foram determinados os indicadores de estresse (concentração plasmática de cortisol, de glicose e número de eritrócitos) e os indicadores imunológicos (atividade respiratória de leucócitos, do sistema complemento - ACH50 - e concentração sérica de lisozima). O CL diminuiu significativamente a concentração plasmática de cortisol em uma (500 mg kg-1) e 3 h (300 mg kg-1) após a inoculação bacteriana, em relação aos peixes do grupo controle. O CL não influenciou a glicemia, o número de eritrócitos e a concentração sérica da lisozima. O CL (100 mg kg-1) aumentou a ARL em 1h e também a ACH50 em 3 h e em todos os tratamentos de CL, 24 h após a inoculação. Os resultados deste estudo demonstram que a exposição aérea e a infecção bacteriana foram estressores para o pacu, causando aumento da glicemia e prejuízo da resposta imune da lisozima e que o CL foi eficiente para reduzir o cortisol e aumentar a atividade fagocítica dos macrófagos e a ACH50 nos peixes. Durante a amostragem inicial, para analisar a eficácia do cloridrato de levamisol como antiparasitário, quatro peixes do grupo controle e de cada tratamento (n=20) foram eutanasiados por secção medular e encaminhados para análise parasitológica.
