Análise histomorfológica e molecular de lesões granulomatosas sugestivas de tuberculose bovina

Abstract

tuberculose bovina é uma enfermidade infectocontagiosa granulomatosa crônica de carácter zoonótico causada pelo Mycobacterium bovis. Essa bactéria pertence ao complexo Mycobacterium tuberculosis que corresponde a um grupo de micobactérias filogeneticamente relacionadas, que apresentam considerável dificuldade no diagnóstico direto em amostras teciduais. O objetivo do presente trabalho foi avaliar linfonodos por meio de diagnóstico histopatológico e molecular, visando à detecção direta do agente em tecido. Durante a linha de inspeção de abate em frigorífico, foram selecionados 150 linfonodos com lesões macroscópicas sugestivas de tuberculose. Na análise histológica pela coloração de Hematoxilia-Eosina (HE), 100% das amostras apresentaram lesões microscópicas compatíveis com tuberculose. A coloração de Zielh-Neelsen (ZN) evidenciou a presença de bacilos álcool ácido resistentes (BAAR) em 6 (4%) amostras. O método de extração para a análise molecular apresentou bons resultados, mas a amplificação dos fragmentos de DNA dos genes hsp65 e IS6110 resultaram em baixa sensibilidade. Contudo a reamplificação do gene IS6110, resultou em positividade de 63,3%, indicando que a dificuldade do diagnóstico molecular em amostras teciduais esta relacionada a quantidade reduzida de bactérias presentes nas amostras. Bovine tuberculosis is a chronic granulomatous infectious disease of zoonotic nature caused by Mycobacterium bovis. This bacterium belongs to the Mycobacterium tuberculosis complex which corresponds to a group of phylogenetically related mycobacteria, which have considerable difficulty in diagnosing directly in tissue samples. The objective of this study was to evaluate lymph nodes through histopathologic diagnosis and molecular targeting the direct detection of the agent in tissue. During the inspection line slaughter in a fridge, we selected 150 nodes with macroscopic lesions suggestive of tuberculosis. In histological analysis by staining Hematoxilia-eosin (HE), 100% of the samples had microscopic lesions compatible with tuberculosis. The coloring Zielh-Neelsen (ZN) revealed the presence of acid- resistant bacilli (AFB) in 6 (4%) samples. The extraction method for molecular analysis showed good results, but the amplification of DNA fragments of hsp65 genes and IS6110 resulted in low sensitivity. However, the reamplification of IS6110 gene resulted in 63.3% positivity, indicating the difficulty of tissue samples for molecular diagnosis related to this reduced amount of bacteria present in samples
