Efeito da sazonalidade sobre a função testicular de cães

Abstract

In temperate zones, seasonal influence on reproductive performance of male dogs has been described, but in tropical conditions it hasn't been studied. The objective of this work was to evaluate the influence of seasonality on testis function. In the study 8 adult dogs were used, with age varying between 1.2 and 6 years old, from 3 breeds (1 Blood Hound, 2 Golden Retrivier, 4 Springer Spaniel). These dogs were active studs, and they were housed in the same cannel, during 14 months. Ejaculates were collected each 15 days. Immediately after the collection, spermatic progressive motility, progressive velocity, pH and sperm morphology, were evaluated. The samples were centrifuged, the pellet re-suspended and frozen, and supernatant was evaluated for total protein, cloride, potassium, sodium, calcium and magnesium. Ultrasonic measurements of canine testis and prostate were performed. Electrophoresis was performed using 12% and 18% SDS-PAGE. The electrophoresis gel was stained with Coomasie blue brilliant R-250, scanned and analysed by a software. Molecular weight was estimated based in a standard weight. Blood collections were performed every 15 days at morning (between 8:30 and 11:30 a.m). Testosterone serum concentrations were determined by solid phase RIA (radioimmunoassay). The data from environmental temperature and pluvial index were obtained from CIIAGRO. Testosterone mean concentrations varied from 1.31 ng/mL to 2.02 ng/mL. The temperature varied from 10.2º C to 32.8º C and the pluvial index was 33 mm3 to 476 mm3. Notable individual variation was observed on testosterone serum concentrations among dogs in all seasons. There was a remarkable decrease on testosterone levels in all animals during Summer, when temperature and humidity were high. Em climas temperados, variações sazonais na reprodução em cães têm sido reportadas, entretanto, em condições tropicais essa influência não tem sido estudada. O objetivo desse trabalho foi avaliar se as modificações ambientais, da Região Sudeste do Brasil, exercem efeitos sobre a função testicular. Neste estudo foram utilizados 8 cães adultos, com idade entre 1,2 a 6 anos, de três raças (1 Blood Hound, 2 Golden Retrivier, 4 Springer Spaniel), em atividade reprodutiva, de um mesmo canil, num período de 14 meses. Colheitas de sêmen foram realizadas quinzenalmente. O ejaculado foi imediatamente avaliado quanto à motilidade e vigor espermático, pH, concentração e morfologia dos espermatozóides. Após a centrifugação, o pellet foi ressuspendido e submetido a congelação. No plasma seminal foram determinadas as concentrações de proteínas totais, sódio, potássio, cálcio, magnésio e cloretos totais. A eletroforese SDS-PAGE foi realizada utilizando-se géis de poliacrilamida nas concentrações de 12 e 18%. Os géis foram corados com Coomasie azul brilhante R-250, as imagens digitalizadas e os pesos moleculares e a densidade óptica integrada estimada. Amostras de sangue foram obtidas a cada 15 dias, sempre no período da manhã, entre 8:30 e 11:30 h. As dosagens séricas de testosterona total foram determinadas por radioimunoensaio (RIA) em fase sólida. Foram realizadas mensurações de próstata e testículos. Os dados referentes às variações de temperatura ambiente (°C) e índice pluviométrico (mm3) foram obtidos no CIIAGRO. A variação de temperatura, nesse período, foi de 9ºC a 32,8ºC, e o índice pluviométrico de 26 mm3 a 476 mm3. A concentração média de testosterona nos animais foi de 131,65 ng/dL a 213,17 ng/dL, com uma grande variação individual.
