Variabilidade intraespecifica de estirilpironas em quimiotipos de Cryptocarya mandioccana: avaliacao da producao em diferentes orgaos e em extratos enzimaticos

Abstract

Cryptocarya mandioccana apresenta quimiotipos caracterizados pela diferença quali e quantitativa na capacidade de produzir estirilpironas de cadeias policetídicas de vários comprimentos. Por isso foi selecionada como modelo para estudos biossintéticos e ecofisiológicos, visando o entendimento da formação e função desses metabólitos secundários. Um método cromatográfico para análise simultânea dos principais metabólitos encontrados nas folhas de C. mandioccana foi desenvolvido. O método foi validado com base nas normas da ANVISA e os parâmetros de seletividade, linearidade, precisão, limite de detecção, limite de quantificação, exatidão e estabilidade ficaram dentro dos critérios de aceitação. Esse método foi utilizado para comparar o perfil metabólico dos órgãos de indivíduos dos quimiotipos F e FS. No perfil de brotos, folhas novas e folhas velhas dos dois quimiotipos foi observado o acúmulo de alcalóides e flavonóides nas folhas velhas, e de substâncias, ainda não identificadas, nos brotos. Estirilpironas foram observadas nas folhas novas do quimiotipo FS e no mesmo órgão do quimiotipo F foi detectada a presença de substâncias ainda não identificadas. No perfil de folhas, galhos, cascas do tronco e raízes dos dois quimiotipos foi observado o acúmulo de alcalóides nas cascas do tronco e raízes, e de flavonóides nas folhas. Estirilpironas foram observadas nas folhas e em baixas concentrações nos galhos e raízes do quimiotipo FS. Experimentos biossintéticos realizados com folhas de plântulas de C. mandioccana confirmaram a atividade da enzima PAL, que converteu L-fenilalanina em ácido trans-cinâmico. No entanto, nas condições experimentais utilizadas para avaliar a atividade da enzima SPS em folhas das mesmas plântulas não foi detectada a formação de estirilpironas.

Cryptocarya mandioccana presents chemotypes characterized by difference in the quali and quantitative capacity of producing styrylpyrones of polyketide chain of various sizes. For that reason, it was selected as model to biosynthetic and ecophysiologic studies, aiming the acquaintance of formation and function of these secondary metabolites. A chromatographic method for simultaneous analysis of the main metabolites found in the leaves from C. mandioccana was developed. The method was validated based on ANVISA regulations and its parameters of selectivity, linearity, precision, detection limit, quantification limit, accuracy and stability showed good results. This method was utilized to compare metabolic profile of the organs of individuals of the F and FS chemotypes. In the profile of sprout, young leaves and old leaves of both chemotypes it was observed the presence of alkaloids and flavonoids in old leaves, and a compound still not identified, in the sprout. Styrylpyrones were observed in the young leaves of the FS chemotype and in the same organ of the F chemotype it was observed the presence of a compound still not identified. In the profile of leaves, branches, trunk barks and roots of both chemotypes it was observed the presence of alkaloids in the trunk barks and roots, and flavonoids in the leaves. Styrylpyrones were observed in the leaves and in low concentration in the brunches and roots of the FS chemotype. Biosynthetic test performed in leaves of seedlings from C. mandioccana confirmed PAL enzyme activity that converted Lphenylalanine in cinnamic acid. However, in the test conditions used to evaluate SPS enzyme activity in leaves of the same seedlings it was not observed the formation of styrylpyrones.
