Abstract

O mecanismo da cardiotoxicidade induzida pela doxorrubicina (DOX) tem sido freqüente objeto de pesquisa. Dentre os vários mecanismos, destaca-se o estresse oxidativo. O objetivo do estudo foi avaliar o efeito de antioxidantes (a-tocoferol e do licopeno), isolados ou associados, na cardiotoxicidade induzida pela DOX. Materias e Métodos - Ratos machos Wistar (n, 132 ratos) foram divididos aleatoriamente em 8 grupos: G1 (controle), G2 (licopeno), G3 (a-tocoferol), G4 (licopeno + a-tocoferol), G5 (DOX), G6 (DOX + licopeno), G7 (DOX + a- tocoferol) e G8 (DOX + licopeno + a-tocoferol). Os animais foram tratados via gavagem com licopeno [(4 mg/Kg/dia) G2, G4, G6, G8], a-tocoferol [(250 mg/kg/dia) G3, G4, G7, G8] e óleo de milho [(volume apropriado) G1, G5] por 7 semanas. Eles também foram tratados intraperitonealmente com DOX [(4 mg/Kg/semana), dose cumulativa de 16mg/Kg] na 3ª, 4ª, 5ª, e 6ª semana e na 7ª semana eles foram sacrificados para coleta de dados após a avaliação ecocardiográfica. Foram também analisados: peso corpóreo, peso cardíaco, cardio-histologia, níveis cardíacos de a-tocoferol e licopeno por HPLC e níveis séricos de colesterol e LDL-colesterol. Doxorubicin is an excellent chemotherapeutic agent utilized for several types of cancer but the irreversible doxorubicin-induced cardiac damage is the major limitation for its use. Oxidative stress seems to be associated with some phase of the toxicity mechanism process. The aim of this study was to evaluate the effect of antioxidants (a-tocopherol and lycopene) alone or in association on doxorubicin-induced cardiotoxicity. Materials and Methods – 132 male Wistar rats were randomly distributed into eight groups: G1 (control), G2 (lycopene), G3 (a-tocopherol), G4 (lycopene + a- tocopherol), G5 (DOX), G6 (DOX + lycopene), G7 (DOX + a-tocopherol) or G8 (DOX + lycopene + a-tocopherol). The animals received lycopene [(4 mg/Kg/day) G2, G4, G6 and G8)], a-tocopherol [(250 mg/kg/day) G3, G4, G7 and G8)] and corn oil [(appropriate volume) G1 and G5)] by gavage for a 7- week period.
