Citotoxicidade, genotoxicidade e expressão gênica em células da polpa dental humana sensibilizadas por cimentos endodônticos

Abstract

O sucesso dos tratamentos endodônticos está relacionado ao correto diagnóstico das alterações pulpares, à qualidade do preparo biomecânico e da obturação dos canais radiculares, bem como à compatibilidade entre os cimentos obturadores com os tecidos adjacentes ao canal. Atualmente a busca de bom comportamento biológico destes cimentos se faz de extrema importância. O presente estudo teve como objetivo analisar a citotoxicidade e a genotoxicidade dos extratos de três cimentos endodônticos e seus efeitos sobre a expressão dos genes BMP2, ALP, RUNX2, BGLAP, OPN e DMP1 em células da polpa dental humana. Foram utilizadas células primárias de tecidos pulpares de terceiro molar com rizogênese incompleta. Cimentos endodônticos foram depositados em placas de 24 poços e armazenados a 37°C por 6 h; cada espécime foi colocado em contato com 2,7 ml de meio de cultura (DMEM-F12) e as amostras foram incubadas por 24 h, a 37°C. Extratos originais (1:1) de cada material foram diluídos em série até 1:32 e analisados quanto a citotoxicidade pelo ensaio de MTT. Após a obtenção de uma diluição que não fosse extremamente tóxica e não interferisse demasiadamente na replicação celular (subdose), foi realizado o teste de micronúcleo para avaliação da genotoxicidade dos materiais. A expressão dos genes foi verificada por RT-PCR em tempo real, em 6, 12 e 24 h. O RNA total foi isolado e retrotranscrito. Os testes de citotoxicidade indicaram que a sobrevivência celular foi afetada pelos extratos na seguinte ordem: EndoREZ > AH Plus > RoekoSeal ≥ MTA. O EndoREZ propiciou o maior número de micronúleos no ensaio de genotoxicidade. A quantificação relativa indicou que em 24 h, o AH Plus e o EndoREZ agiram de modo similar, aumentando a expressão da OPN e reduzindo a ALP; o RoekoSeal gerou expressão da OPN...

The success of endodontic therapy is related to the correct pulp and periapical alterations diagnosis; to the good biomechanical instrumentation and to the root canal sealing quality; to the compatibility between the endodontic sealers and the root canal surrounding tissues as well. Nowadays, the seeking for a sealer with good biological behavior is extremely important. The aim of this study was to analyze the cytotoxicity and the genotoxicity of three endodontic sealers and their effects on the expression of the genes BMP-2, ALP, RUNX2, BGLAP, SPP1 and DMP1 in human dental pulp cells. Primary cells from the 5th passage obtained from pulp tissue of third molar with incomplete rizogenesis were used in this study. Endodontic sealers were placed in 24 well plates and stored at 37°C for 6 hours. After that, each specimen was covered with 2.7 ml of cell culture media (DMEM-F12) and incubated afterwards at 37°C for 24 h. Original extracts (1:1) of each material were serially diluted up to 1:32 and the citotoxicity was analyzed by MTT assay. After obtaining extracts subdose, non very toxic or non interfering on cell replication, genotoxicity was evaluated by micronucleus test. The gene expression was accessed by Real Time RT-PCR, after extracts exposure of 6, 12 and 24 h. Total RNA was isolate and retrotranscribed. The citotoxicity test has showed that cell survival was affected in the following order: EndoREZ > AH Plus > Roeko Seal ≥ MTA. EndoREZ has caused the highest score in micronucleus assay.
