Abstract

A alteração biológica mais evidente observada durante o exercício físico é o aumento da taxa metabólica, com elevada demanda de oxigênio, que pode aumentar a formação de espécies reativas de oxigênio (ROS) potencialmente danosas ao organismo, ocorrendo lipoperoxidação como conseqüência. Foi desenvolvido um método de isolamento simultâneo de células mononucleares e polimorfonucleares de sangue periférico eqüino, utilizando gradiente descontínuo de ficoll-hipaque d=1,077 e d=1,119. A separação de ambos os tipos celulares resultou em alto grau de pureza e viabilidade. Foram avaliadas funções de monócitos e polimorfonucleares em eqüinos da raça Árabe suplementados com vitamina E comparando-se com um grupo controle em duas provas de exercício progressivo em esteira de alta velocidade antes e depois de serem submetidos a um protocolo de treinamento. Os resultados indicam que o treinamento e a suplementação com vitamina E preservaram as funções dos monócitos eqüinos quanto à geração de anion superóxido (O2 -), peróxido de hidrogênio (H2O2), óxido nítrico (NO) e fator de necrose tumoral alfa (TNF-a) ao mesmo tempo em que preveniram a produção exacerbada dessas substâncias. Enquanto as produções de H2O2 e O2 - por polimorfonucleares pelo grupo de animais suplementados foram mais elevadas. Isso indica a exacerbação desta função dos polimorfonucleares dos eqüinos pela vitamina E antes e após a prova de exercício físico em esteira de alta velocidade. A vitamina E suplementar na dieta promoveu menor formação de malondialdeído (MDA), ou seja, ocorreu atenuação da lesão oxidativa.

The clearest biological alteration observed during physical exercise is the increase of metabolic rate, with a high oxygen demand. It can increase the reactive oxygen species (ROS) formation potentially harmful to the organism, occurring lipoperoxidation as a result. It was developed a method of simultaneous separation of peripheral blood mononuclear and polymorphonuclear cells in horse utilizing discontinuous Ficoll-Hypaque gradients d=1,077 and d=1,119. Both cells types isolation resulted in high purity and viability degree. It was evaluated functions in monocytes and polymorphonuclear cells in Arabian horses supplemented with vitamin E by comparing a control group in two incremental treadmill exercises before and after been submitted to a training protocol. Results indicated that training and vitamin E supplementation preserved the monocyte functions regarding superoxide anion generation (O2 -), hydrogen peroxide (H2O2), nitric oxide (NO) and tumor necrosis factor alpha (TNF-a) at the same time that they prevented an excessive production of those substances. Whereas H2O2 e O2 - productions by polymorphonuclear cells in supplemented animals group were higher. It indicates an exacerbation of that horse polymorphonuclear cells function by vitamin E before and after incremental treadmill exercises. Supplementary vitamin E in the diet promoted a lower malondialdeide (MDA) formation, namely occurred attenuation in oxidative stress.
