Abstract

El principal objetivo de este trabajo es estudiar el mecanismo por el cual la síntesis de proteínas disminuye durante el envejecimiento. Durante el proceso de envejecimiento se produce una disminución significativa de la síntesis de proteínas en todos los órganos de los distintos organismos estudiados. Sin embargo, hasta el momento no se sabe ni el mecanismo por el que esto ocurre ni que etapa de la síntesis de proteínas se encuentra especialmente afectada. Considerando la principal teoría del envejecimiento, que lo relaciona con el acumulo del daño oxidativo, se ha determinado en primer lugar qué etapa de la síntesis de proteínas (iniciación, elongación o terminación) podría estar más afectada por el envejecimiento. Esto se ha estudiado mediante la determinación de perfiles de polisomas y mediante la determinación del tiempo de terminación de cadenas nacientes de proteínas de tamaño medio. Nuestros resultados ponen de manifiesto que es la etapa de elongación la más afectada. En segundo lugar, se ha estudiado el mecanismo por el que el envejecimiento afecta la etapa de elongación. Para ello, nos hemos centrado en la proteína más relevante en esta etapa, el factor de elongación 2 (EF-2)(Riis y Rattan, 1990) estudiando a nivel molecular las modificaciones post-traduccionales más importantes que puede sufrir esta proteína: oxidación y la ADP-ribosilación. Nuestros resultados ponen de manifiesto que el EF-2 hepático de ratas viejas presenta un mayor grado de daño oxidativo, una menor actividad en término de cantidad de diftamida y un mayor grado de fragmentación, lo cual podría justificar el enlentecimiento de la fase de elongación observada en estos animales viejos. El hecho de que a nivel molecular las modificaciones en el EF-2 sean similares en ratas viejas y ratas jóvenes tratadas con compuestos oxidantes sugiere que los radicales libres pueden estar implicados en la inhibición de la síntesis
