Abstract

Las cianobacterias crecenprincipalmente en aguasdulcesde zonas tropicales y subtropicales, aunque cada vez resulta más evidente su presencia en aguas dezonas templadas. Algunas especies de cianobacterias son capaces de producir cianotoxinas, siendo la microcistina-LR (MC-LR) y la cilindrospermopsina (CYN) dos de las que despiertan mayor interésdesde el punto de vista toxicológico.Se ha descrito que MC-LR es hepatotóxica y CYN citotóxica. Sin embargo,existen algunosestudios que muestran la capacidad de ambas parainducir efectos neurotóxicos. Porello, el objetivo del presente trabajo fue estudiar el posible efecto queMC-LR y CYNpudieran tener, tanto de forma aislada comoen combinación, sobre lalínea celular de neuroblastoma humanoSH-SY5Y. Para ello se realizaron los ensayos de citotoxicidad basal de captación de rojo neutro y reducción de la sal de tetrazolio MTS. Tras exponerlas células a 0-100 μg/mL de MC-LR y 0-10 μg/mL de CYNdurante 24 y 48 horas, se observó que ambas cianotoxinas producíanefecto neurotóxico, resultando CYN más citotóxica que MC-LR. Para estudiarlos efectos de su combinación se emplearon la concentración efectiva media de cada cianotoxina y fracciones de éstas. Así, este estudio mostróque a concentraciones elevadas, éstaresultabamás citotóxica quelas cianotoxinas de manera individual. Además,paraevaluar el tipo de interacción que se producíaentre MC-LR y CYN,se empleóel método de isobologramas, por el que es posible determinar si una combinación es antagónica, aditiva o sinérgica. Tras analizar los resultados, se dedujoque la combinaciónde MC-LR y CYN erade tipo antagónico, si bien se observó una tendenciahaciaaditividad aconcentraciones elevadas. En base a estosresultados, se demostró que las cianotoxinas MC-LR y CYN presentan capacidad neurotóxicaen la línea celular SH-SY5H, tantoaisladas comoen combinación
