Abstract

Esta revisión bibliográfica trata de explicar de forma sencilla qué son los inhibidores de la caspasa 8. Para ello se explicará en principio qué es la muerte celular, qué tipos de muerte celular se llevan a cabo en la célula, la vía de muerte celular en la que está implicada esta enzima, otros procesos en los que se encuentra implicada no relacionados con la muerte celular para así entender su importancia y consecuencias de su inhibición. Además, se describirá qué son las caspasas, qué tipos hay, cómo es su mecanismo de acción o cuál es su estructura para poder así entender de una forma más sencilla los diferentes métodos que podemos utilizar para inhibirla. Principalmente hemos dividido los tipos de inhibidores en tres grandes grupos: (1) inhibidores de tipo microARN, tanto los que encontramos en el propio organismo como aquellos que podemos sintetizar, junto con algunos de los ejemplos más relevantes que se han utilizado en investigación; (2) los ratones KO o doble KO con el gen de la caspasa 8 delecionado; y por último (3) los inhibidores sintéticos de caspasa 8, incluyendo las modificaciones más importantes que se le hacen para mejorarlos. De cada tipo se explicará cual es el mecanismo de acción llevado a cabo por cada uno de ellos al igual que otros complejos que también participan en el proceso. Finalmente se expondrán algunas de las ventajas e inconvenientes de cada tipo de inhibidor. No obstante, es un tema difícil de abarcar que aún se encuentra en investigación.

This review will focus on the different caspase 8 inhibitors. To achieve this objective, we will start describing the cellular death processes as well as the cellular death pathways in which the caspase 8 is involved. In addition, the implication of caspase 8 in other processes will be discussed. Focusing on the different caspase types, structure, and mechanism of action, will help to find the optimal method to inhibit this important type of enzymes. The mechanisms to inhibit or avoid the action of caspase enzymes can be divided into three main categories: (1) microRNA type inhibitors (biological and synthetic) that will 3 be discussed along with some of the most relevant examples that have been used in research, (2) KO mice or double KO mice, with a deletion of the caspase 8 gene, and (3) synthetic caspase 8 inhibitors. The different mechanism of action of each type of inhibitor will be explained as well as the main advantages and disadvantages. Finally, it is important to note, that the topic of the present review is nowadays under intense investigation and it is presumable that in the next years a lot of new studies will provide important information regarding the optimal strategy to inhibit the caspase 8 and its therapeutical benefit.
