¿Qué efecto tiene la inclusión de Hidroxipropil-betaciclodextrina (HPβCD) en la formulación de liposomas de HPβCD-acetazolamida?

Abstract

En el presente trabajo de investigación, se ha querido estudiar los efectos que tendría la inclusión de la hidroxipropil-beta-ciclodextrina en una formulación de liposomas de acetazolamida, un inhibidor de la anhidrasa carbónica utilizado para el tratamiento del glaucoma. Acetazolamida es utilizada por vía oral en el tratamiento del glaucoma, presentando aceptables valores de disminución de presión intraocular. Sin embargo, requiere una posologia compleja, y los estudios realizados en los últimos anos han demostrado que la administración tópica ocular presenta mucho mejores resultados y una mejoría notable en esta patología. El principal problema que presenta este tipo de formulaciones para este fármaco reside en su baja biodisponibilidad tras la administración tópica ocular, debido a su baja solubilidad acuosa y permeabilidad. En base a ello, se han empleado distintas estrategias solubilizantes que ayuden a mejorar estas propiedades. Entre estas, se ha seleccionado el uso de ciclodextrinas, que son moléculas complejantes que aumentan la solubilidad acuosa del fármaco. A su vez, introducir este complejo en un sistema vesicular como son los liposomas, podría constituir una buena vía para mejorar la estabilidad del complejo, aumentar el tiempo de contacto de la formulación con la superficie ocular y mejorar las características de permeabilidad del fármaco
