Neuroprotección en un nuevo modelo de animal transgénico

Abstract

El objetivo general de este proyecto fue analizar en un modelo de parkinsonismo experimental en ratones, si el aumento del poder reductor de forma selectiva en las neuronas dopaminérgicas nigrostriatales (neuronas DNS9, mediante la sobre-expresión del enzima glucosa-6-fosfato deshidrogenada (G6PD), les confiere protección. Además nos interesó estudiar algunos aspectos relacionados con la regulación de la expresión de dicho enzima en células excitables. Los objetivos específicos de este trabajo fueron: 1. Generar un animal transgénico que sobre-exprese la G6PD en las neuronas DNS utilizando un promotor específico (promotor de la tirosina hidroxilasa) de células catecolaminérgica. 2. Caracterizar los animales transgénicos. Comprobar mediante técnicas histológicas y de actividad enzimática, si la sobre-expresión del transgén es específica y funcional. 3. Demostrar que la sobre-expresión de la G6PD no afecta a los niveles de otras proteínas ni a parámetros fisiológicos del animal como tiempo de vida, peso, capacidad reproductiva, etc. 4. Analizar la degeneración de la vía nigrostriatal producida por la neurotoxina MPTP en los ratones transgénicos, mediante técnicas densitométricas, amperométricas, bioquímicas y de biología molecular. 5. Estudiar la toxicidad del MPTP en los animales transgénicos utilizando modelos crónicos de administración de dicha sustancia, mimetizando de este modo la neurodegeneración progresiva que ocurre en la enfermedad de Parkinson. 6. Caracterizar la neurotoxicidad producida por la administración aguda de MPTP en animales transgénicos de edad avanzada (2-2,5 años). 7. Estudiar la regulación de la expresión génica de la G6PD por la hipoxia crónica en células PC12.
