Abstract

Nuestro objetivo es utilizar la diaminobutanona y los mutantes de Phycomyces resistentes a este inhibidor para investigar el papel de las poliaminas. Un aspecto de este trabajo, puramente descriptivo, investiga las variaciones cuantitativas de las poliaminas y la descarboxilasa de la o ... rnitina a lo largo de los ciclos de vida, sexual y asexual, de la estirpe silvestre. Por otra parte nos proponemos caracterizar los mutantes resistentes a diamonobutanona, estudiando los aspectos químicos, enzimáticos, genéticos y fisiológicos. Un aspecto derivado del análisis de la dominancia y necesidad de las mutaciones spe nos llevó a un problema muy poco estudiado en los organismos cenocíticos. Estos organismos no tienen tabicaciones (septos) que dividan una célula de otra y sus núcleos pueden desplazarse por el micelio. ¿Variará la proporción de un tipo de núcleos dentro de un heterocarionte ejerciendo una presión selectiva sobre este?Las poliaminas son compuestos nitrogenados policatiónicos de bajo peso molecular que se encuentran en todos los organismos. Son esenciales para el crecimiento y tienen papeles fisiológicos importantes, pero desconocidos.Se han investigado las variaciones en las concentraciones de poliaminas a lo largo de los ciclos de vida del hongo Phycomyces blakesleeanus, que tiene putrescina y espermidina, pero no espermina. Se han utilizado cuatro mutantes (spe) residentes a diaminobutanona, un analogo sintetico de la putrescina que inhibe a la descarboxilasa de la ortinina.En Phycomyces se ha visto una relación entre la máxima concentración d putrescina y la fase de crecimiento exponencial.Se ha secuenciado un fragmento del gen para la descarboxilasa de la ornitina de Phycomyces, al que llamamos speA, cuya secuencia de aminoácidos es muy semejante a la de Mucor.Las mutaciones spe no afectan de manera especial a ningun paso del desarrollo.
