Abstract

En esta Tesis se ha estudiado la actividad carotenogénica de los extractos celulares de la estirpe silvestre y distintos mutantes de Phycomyces, así como el efecto sobre dicha actividad de ciertos factores. También se ha aislado y caracterizado un nuevo mutante en ... la carotenogénesis de dicho organismo. Se concluye que:- La carotenogénesis in vitro se ve afectada por el nivel del oxígeno en el ensayo. La síntesis de fitoeno es independiente de la presencia de oxígeno, siendo este requerido para la desaturación de fitoeno hasta licopeno.- Las mutaciones en los genes carB y carR provocan la inactividad de las enzimas deshidrogenasa del fitoeno y ciclasa del licopeno, respectivamente.- La estirpe C2, que tiene una mutación en el gen carA, presenta una actividad carotenogénica in vitro significativamente menor que la estirpe silvestre.- Se ha encontrado que el fitoeno trans participa en la ruta biosintética de carotenos en Phycomyces. - El retinol estimula la actividad de las enzimas carotenogénicas.- La difenilamina inhibe la actividad de la enzima deshidrogenasa del fitoeno y, a la vez, provoca una variación en la acumulación de isómeros de ese caroteno.- Se ha aislado una estirpe mutante de Phycomyces (S3o3) que acumula ß-caroteno de carotenoproteína.- La producción de carotenoproteína está determinada por un factor genético extranuclear y puede requerir también la presencia de una mutación en el gen nuclear carS presente, originalmente, en la estirpe S3O3.- La iluminación o presencia de retinol en los medios de cultivo inhiben la producción del complejo carotenoproteínico de la estirpe S3O3.- En la inhibición de la producción de carotenoproteína por el retinol juega un papel importante el anillo de esta molécula, siendo poco relevante la naturaleza del grupo funcional presente en el extremo de la cadena lateral.|
