Penetración y actividad intracelular de cinco fluorquinolonas en leucocitos polimorfonucleares humanos

Abstract

La finalidad de este estudio ha sido determinar la capacidad de un grupo de quinolonas; fluorquinolonas, de penetrar en el interior de los leucocitos polimorfonucleares humanos, estudiar los mecanismos involucrados en dicha penetración y su posible actividad intracelular contra determinadas bacterias. Lo realmente importante y determinante a la hora de establecer una terapéutica antimicrobiana en infecciones intracelulares es que el antibiótico se concentre y permanezca activo en el lugar donde se localiza la bacteria (y que las condiciones físico-químicas reinantes no influyan negativamente en su actividad intracelular). - Norfloxacina, ofloxacina, levofloxacina, lomefloxacina y fluoxacina alcanzan concentraciones intracelulares en los pmn humanos de 3 a 5 veces superior a las extracelulares. - La penetración intracelular de estas fluorquinolonas depende de la temperatura y pK extracelular, pero no de la viabilidad celular. losinhibidores metabólicos tampoco parecen influir negativamente en dicha penetración. - Ninguna de las fluorquinolonas evaluadas afecto a la producción de peróxido de hidrogeno y radicales superóxido por los pmn humanos, mostrándose todas activas en reducir la supervivencia intracelular de S. aureus en pmn humanos.
