El secoiridoide del aceite de oliva virgen extra oleocantal como inhibidor de la osteoclastogénesis

Abstract

Un desequilibrio en la homeostasis ósea puede suponer la aparición de ciertas patologías como la osteoporosis (OP), artritis reumatoide (AR), metástasis óseas líticas y enfermedad ósea de Paget. Oleocantal (OLE), un secoiridoide característico y exclusivo de la familia Oleaceae, se encuentra principalmente en el aceite de oliva virgen extra (AOVE). Estudios previos han demostrado sus efectos antioxidantes, antiinflamatorios, antimicrobianos, anticancerígenos y neuroprotectores. El objetivo de este estudio ha sido estudiar el funcionalismo de este secoiridoide en un modelo de osteoclastogénesis inducida por M-CSF y RANKL a partir de células de médula ósea de ratón y dilucidar los posibles mecanismos moleculares involucrados. Los macrófagos de la médula ósea fueron aislados de los huesos de ratones CD1 y estimulados con M-CSF/RANKL en presencia o ausencia del tratamiento con OLE (25, 50 y 100 μM). Tras el tratamiento, se realizaron ensayos de viabilidad celular (SRB), tinción y actividad TRAP, determinación de los niveles de citoquinas (IL-6 y TNF-α) y expresión de genes marcadores de osteoclastos (NFATc1, TRAP, MMP-9 y CTSK) mediante qPCR. Los resultados obtenidos muestran que el tratamiento con OLE no afecta a la viabilidad celular a las concentraciones ensayadas (25, 50 y 100 μM). OLE fue capaz de reducir significativamente el número de células multinucleadas TRAP-positivas y la actividad de dicha enzima. El tratamiento con el secoiridoide (50 y 100 μM), produjo una disminución significativa de los niveles de IL-6 y TNF-α. Además, pudimos observar que OLE reguló la expresión genética de NFATc1, TRAP, MMP-9 y CTSK. Estos resultados nos permiten concluir que OLE podría formar parte de nuevos tratamientos destinados a patologías óseas con exacerbación de la osteoclastogénesis por inhibir la diferenciación de osteoclastos junto a su capacidad antiinflamatoria.
