Sensores de ARN de cadena doble en la inmunidad innata

Abstract

La inmunidad innata es uno de los mecanismos principales en la defensa contra la infección viral. La detección de dsARN viral es uno de los instrumentos que tenemos para combatir la infección. Este dsARN es reconocido por las proteínas de unión a dsARN que distinguen entre el dsARN exógeno y el endógeno gracias a ciertas características que presenta el dsARN viral. Entre estas proteínas encontramos a la familia RLR (RIG-I, MDA5 y LGP2) que se encarga de activar una cascada de señalización que termina con la producción de IFN tipo I, citoquinas, inflamación e ISGs. Otras proteínas importantes son PKR (inhibición de la traducción viral), ADARs (edición del ARN, tanto viral como endógeno), OASes (supresión de la transcripción), Dicer/Drosha (biogénesis de miARN y siARN; silenciamiento del ARN viral), TRBP/PACT (cofactor en el silenciamiento del ARN) y otras helicasas DExD/H (helicasas DEAD-box y helicasas DEAH-box) que actúan como cofactores en la defensa antiviral y en la activación de RIG-I y MDA5. Estudios sobre la interacción entre algunas de ellas describen un aumento de la actividad antiviral de la proteína potenciada o, al contrario, una inhibición de esta actividad. Esta inhibición se da principalmente cuando el sustrato dsARN es endógeno para evitar cualquier complicación como enfermedades neurodegenerativas (p.ej. síndrome de Aicardi-Goutières), aunque la inhibición de ciertas proteínas también se da cuando existe infección viral. En esta revisión bibliográfica nos vamos a centrar en recoger, de forma resumida, las principales funciones de cada una de las proteínas de unión a dsARN y principales interacciones entre ellas estudiadas hasta el momento.
