Estrategias reparativas en el sistema oculomotor tras la lesión mediante la administración del factor de crecimiento del endotelio vascular (VEGF)

Abstract

El factor de crecimiento del endotelio vascular (VEGF) es una proteína englobada en la familia de los factores tróficos. Descrito en un principio como un factor implicado en el sistema vascular, actualmente se conocen sus efectos en diversos tejidos. De espacial relevancia es el papel que juega en el sistema nervioso, donde su déficit está asociado a enfermedades neurodegenerativas. Ello es que ratones transgénicos con bajos niveles de expresión de VEGF desarrollan una degeneración progresiva de las motoneuronas que junto al debilitamiento muscular conlleva finalmente a la muerte, síntomas similares a los que se dan en la esclerosis lateral amiotrófica (ELA). Sin embargo, se ha observado que la administración de VEGF en este modelo animal produce una reducción en la muerte de las motoneuronas, aumentando la supervivencia y la esperanza de vida. Hasta la fecha se desconoce el estado funcional en el que se encuentran estas motoneuronas ‘rescatadas’ tras la administración de VEGF. Es por ello que el propósito de este trabajo es el estudio de las propiedades de disparo y de los contactos sinápticos de motoneuronas lesionadas a las que se les ha administrado VEGF siguiendo diferentes protocolos temporales y metodologías. Este trabajo de investigación aporta importantes resultados que afianzan el posible potencial papel terapéutico del VEGF en enfermedades neurodegenerativas. Adicionalmente, se caracteriza funcional y morfológicamente los dos tipos de motoneuronas que se encuentran en el núcleo lesionado de nuestro modelo experimental. Este estudio abre nuevas posibilidades a futuros trabajos en los que se investigue como responden ambos tipos de motoneuronas a la lesión y administración de VEGF.
