Abstract

Eurygaster integriceps (Put.) (Süne, Hemiptera: Scutelleridae) ülkemiz tahıl tarlalarında önemli derecede ürün kayıplarına yol açan bir zararlıdır. Günümüze kadar bu zararlıya karşı kimyasal mücadele yapılmasına karşın etkin bir mikrobiyal mücadele yöntemi uygulanamamıştır.Bu çalışmada, E. integriceps (Put.)'in bakteriyal florasının belirlenmesi ve elde edilen bakteri türlerinin E. integriceps (Put.)'e karşı olası biyolojik mücadele etmeni olarak kullanılması amaçlanmıştır. Bu amaçla Batman ilindeki tahıl alanlarından toplanan E. integriceps (Put.) erginleri incelenmiş, zararlının bakteriyal florası belirlenmiş ve bu zararlıya karşı mikrobiyal mücadele potansiyelleri belirlenmiştir.Sonuç olarak, E. integriceps (Put.) erginlerinden 11 bakteri izole edilmiştir. İzole edilen bakterilerin dördü tür seviyesinde, sekizi ise cins seviyesinde tanımlanmıştır. Bu çalışma kapsamında bakterilerin tür tayininde, rutin olarak kullanılan morfolojik, fizyolojik ve biyokimyasal özelliklerin yanı sıra, bakteriler API 20NE ve API Staph panel test sistemleri kullanılarak metabolik enzim profilleri ve biyokimyasal özellikleri belirlenmiştir. Moleküler karakterizasyon çalışmalarında ise 16S rRNA dizin analizi kullanılmıştır.Yapılan karakterizasyon çalışmaları sonrası zararlının bakteriyal florası Pantoea sp. S1, S5, S7, S8, S10, S11, Pantoea agglomerans S2, S3, S4, Pseudomonas sp. S6 ve Micrococcus luteus S9 olarak belirlenmiştir. Dört gün içersinde bakteriyal izolatların E. integriceps (Put.) erginleri üzerindeki ölüm oranları da belirlenmiştir. En yüksek ölüm oranı %100 ile Pantoea sp. S1, Pantoea agglomerans S4 ve Pantoea sp. S7'den elde edilmiştir. Diğer patojenite değerleri ise %55 ile %88 arasında değişmektedir. Test edilen entomopatojenik funguslardan en yüksek ölüm oranı ise %100 ile Isaria fumosorosea ARSEF8356'dan elde edilmiştir. Test edilen diğer fungusların ölüm oranı ise %33 ile %50 arasında değişmektedir. Sonuç olarak, Pantoea sp. S1, Pantoea agglomerans S4, Pantoea sp. S7, Beauveria bassiana ARSEF8356 ve Isaria fumosorosea ARSEF8356 E. integriceps (Put.) ile mücadelede ümit verici görülmektedir.

Eurygaster integriceps (Put.) (Hemiptera: Scutelleridae) is a very serious pest which causes significant crop losses in Turkey?s cereal field. So far, despite of applications of chemical control, a efffective microbial control method has not been implemented against this pest.The aim of this research is to determined the bacterial flora of E. integriceps (Put.) and to use obtained bacterial species against this pest a possible biological control agent. For this purpose, E. integriceps adults collected from cereal fields in Batman were examined, the bacterial flora of this pest and potentials of isolated species as a microbial control agent were determined against this pest.As a result, 11 different bacteria were isolated from E. integriceps (Put.) adults. Four of these bacteria were determined at species level and seven of them were characterized at genus level. In this study, in addition to morphological, physiological and biochemical properties, metabolic enzyme profiles and biochemical properties of these bacreia were determined using API 20E and API Staph panel test systems. Also, 16S rRNA gene sequence analysis was used in molecular characterization studies.Bacterial flora of this pest was identified as Pantoea sp. S1, S5, S7, S8, S10, S11, Pantoea agglomerans S2, S3, S4, Pseudomonas sp. S6 and Micrococcus luteus S9 based on characterization studies. Also, mortality of bacterial isolates against E. integriceps (Put.) adults within four days were determined. The highest mortality was obtained from from Pantoea sp. S1, Pantoea agglomerans S4 and Pantoea sp. S7 with 100%. Also, mortality of other bacterial species ranged from 33% to 88%. The highest mortality among the tested entomopathogenic fungi was obtained from Isaria fumosorosea ARSEF8333 with 100%. Mortality of other fungi change ranged from 33% to 50%. Consequently, Pantoea sp. S1, Pantoea agglomerans S4, Pantoea sp. S7, Beauveria bassiana ARSEF8356 and Isaria fumosorosea ARSEF8356 seem to be promising candidates in control of E. integriceps (Put.).
