Abstract

Allo scopo di rivelare il DNA provirale di Maedi-Visna Virus (MVV) su polmoni ovini con infezione naturale in atto, sono stati utilizzati campioni inclusi in paraffina provenienti da n. 2 pecore positive all’AGID test per Maedi, che presentavano all’esame istologico tipici quadri di polmonite linfoproliferativa interstiziale. Su di essi sono state eseguite prove di ibridazione in situ (i.s.h.) mediante sonda a DNA sintetizzata in PCR, specifica per un segmento di 291 bp del tratto LTR del genoma provirale; la sonda, marcata con digoxygenina, è stata rivelata con sistema antiDIG-AP-NBT. L’i.s.h. ha consentito di rilevare, in entrambi i casi, segnali forti e regolari localizzati in pneumociti II, epiteli bronchiali e macrofagi. Nei primi i segnali sono stati osservati esclusivamente nel nucleo, mentre i macrofagi hanno manifestato aree positive anche in sede citoplasmatica. I risultati ottenuti inducono a ritenere che il virus, attraverso l’integrazione della quota provirale in cellule dotate di capacità moltiplicativa, persegue anche una strategia di amplificazione, oltre che di diversificazione genica imputabile all’ “infedeltà” della sua trascriptasi inversa.
